The 2016 Summer Olympics, officially the Games of the XXXI Olympiad and also known as Rio 2016, was an international multi-sport event held from 5 to 21 August 2016 in Rio de Janeiro, Brazil, with preliminary events in some sports beginning on 3 August. Rio de Janeiro was announced as the host city at the 121st IOC Session in Copenhagen, Denmark, on 2 October 2009.

The flagbearer for the opening ceremony was Andrew Thomas Mlugu, who was Tanzania's first Olympic judoka. The country's participation at Rio de Janeiro marked its sixth consecutive appearance at the Summer Olympics. Three athletes from São Tomé and Princípe were selected for the Games. Romário Leitão and Celma Bonfim da Graça participated in athletics and Buly Triste in flatwater canoeing. Bonfim was the only female on the roster and the only member with prior Olympic experience. Triste was the first male athlete to carry the São Tomé and Princípe flag at the opening ceremony. São Tomé and Princípe has yet to win its first Olympic medal.

The delegation included two track and field athletes, Benjamin Enzema and Reïna-Flor Okori who both qualified for the Games through wildcard places since their fastest times did not meet the required qualification standards, Okori was selected as the flag bearer for the opening and closing ceremonies. Enzema did not advance beyond the qualification round for the men's 800 metres event and Okori was unable to start the women's 100 metre hurdles contest.

This was the nation's twelfth appearance at the Summer Olympics, with the exception of the 1976 Summer Olympics in Montreal and the 1988 Summer Olympics in Seoul, because of the African and North Korean boycott, respectively.

Both qualified for the Games through wildcard places because they did not match the required qualification standards. Karma was selected as the flag bearer for the opening ceremony while Lenchu held it at the closing ceremony. Karma was eliminated at the Round of 64 while Lenchu exited the competition after the shooting qualification round.
The Gambia, officially the Republic of the Gambia, competed at the 2016 Summer Olympics in Rio de Janeiro, which was held from 5 to 21 August 2016. The country's participation at Rio marked its ninth appearance at the Summer Olympic Games since its début at the 1984 Summer Olympics. The delegation included two track and field athletes, Adama Jammeh and Gina Bass, who both qualified after meeting the qualification standards for their respective events, one judoka, Faye Njie, who made the Games through a quota place and one swimmer, Pap Jonga, who earned a universality place to enter the Games. The Gambia made their début appearances in the judo and swimming events. Bass was selected as the flag bearer for the opening and closing ceremonies. All four athletes were eliminated from the first rounds of their events.
Carmen Patricia Martínez is a Paraguayan long distance runner who specialises in the marathon. She finished 115th in the 2016 Olympic marathon in a time of 2 hours 56 minutes and 43 seconds. She served as the flag bearer for Paraguay during the closing ceremony.
Ángela Melania Castro Chirivechz is a Bolivian race walker. She competed in the women's 20 kilometres walk event at the 2016 Summer Olympics finishing in 18th place. It was the best result ever for Bolivia in Olympic athletics. She was the flag bearer for Bolivia during both the opening ceremony and closing ceremony.
