The county seat and the largest city is Harrisburg, Pennsylvania's state capital and ninth largest city. The county was created on March 4, 1785, from part of Lancaster County and was named after Louis Joseph, Dauphin of France, the first son of King Louis XVI.

Capital Area Transit (CAT), also known as the Cumberland-Dauphin-Harrisburg Transit Authority, is a regional public transportation agency that operates bus and paratransit service in the Harrisburg–Carlisle metropolitan statistical area. Its scheduled route bus service covers much of the southern half of Dauphin County and the eastern half of Cumberland County. It also operates two bus routes into northern York County. CAT's shared ride/paratransit operations serve residents throughout Dauphin County. In 2021, the system had a ridership of 1,128,600.
Penn State Health Milton S. Hershey Medical Center (MSHMC) is a 628-bed non-profit, tertiary, research and academic medical center located in Hershey, Pennsylvania, servicing the Central Pennsylvania area. MSHMC is the region's only university-level academic medical center. The hospital is owned by the Penn State Health System and is the largest hospital in the system. MSHMC is affiliated with the Pennsylvania State University College of Medicine. MSHMC is also an ACS designated level I adult and pediatric trauma center and has a helipad to handle medevac patients. Attached to the medical center is the 122 bed Penn State Children's Hospital, which treats infants, children, adolescents, and young adults up to the age of 21.

Polyclinic Medical Center, also known as Polyclinic Hospital, is a polyclinic in Harrisburg, Pennsylvania, and part of UPMC Pinnacle, a regional system of the University of Pittsburgh Medical Center (UPMC) that serves South Central Pennsylvania. Originally opening in 1909. Most of the building is occupied by Pennsylvania Psychiatric Institute. On other floors are other programs, including two addiction clinics, physical and occupational therapy, audiology and hearing center, HIV clinic, outpatient laboratory, primary care office, and surgery optimization.

The UPMC Hillman Cancer Center (Hillman), previously titled the University of Pittsburgh Cancer Institute (UPCI), is a National Cancer Institute (NCI)-designated Comprehensive Cancer Center located in the Hillman Cancer Center in the Shadyside neighborhood of Pittsburgh, Pennsylvania, United States, adjacent to UPMC Shadyside. The only NCI-designated cancer center in Western Pennsylvania, Hillman is composed of collaborative academic and research efforts between the University of Pittsburgh, the University of Pittsburgh Medical Center (UPMC), and Carnegie Mellon University. Hillman provides clinical cancer care to some 74,000 patients treated at its facilities at both the Hillman Cancer Center location in the Shadyside neighborhood of Pittsburgh and at UPMC-affiliated sites throughout Pennsylvania, New York, Ohio, and overseas locations. Founded in 1984, Hillman became the youngest cancer center in history to achieve NCI-designation. As of 2007, Hillman had received nearly $200 million in funding from the National Cancer Institute, which ranks it as one of the top ten cancer research institutes.

Inter-city service is also provided to Hershey, Pennsylvania. In 2021, the system had a ridership of 213,800, or about 900 per weekday as of the third quarter of 2022.

It is located in Hershey near the Penn State Milton S. Hershey Medical Center and Penn State Children's Hospital, the medical school's principal affiliate. The medical school includes 26 basic science and clinical departments and a broad range of clinical programs conducted at its hospital affiliates and numerous ambulatory care sites in the region.
