Poligeenan (CAS No. 53973-98-1) is a low viscosity, low molecular weight, sulfated polygalactan polymer used exclusively in clinical diagnostic applications. The polymer is derived from the molecular backbone of carrageenan.
The name "poligeenan" was confirmed in 1988 by the United States Adopted Names (USAN) Council. [1] The Council is sponsored by the American Medical Association, the American Pharmaceutical Association, and the US Pharmacopeial Convention Inc.
Prior to 1988, poligeenan was referred to as "degraded carrageenan" in the scientific literature. The terms "poligeenan" and "degraded carrageenan" were often used interchangeably in research articles and reports. [2] [3] The use of the term "degraded carageenan" caused confusion, which resulted in the attribution of the negative observations associated the polymer being attributed to carrageenan. [4] [5] Poligeenan and carrageenan are completely different products.
The primary use for poligeenan is for clinical diagnostic applications, and specifically the suspension of barium sulfate slurries used in X-ray studies of the mouth, throat, and esophagus during swallowing. For this application, poligeenan delivers three essential properties: First, its viscosity imparts lubricity to make the slurry easier to swallow. Second, this same viscosity is high enough to maintain full suspension of all barium sulfate particles during the complete X-ray procedure. Third, the high charge density of the poligeenan solution (ester sulfate units) prevents caking or aggregation of the barium sulfate particles during long-term storage of ready-to-use slurry products, these being easily reconstituted by shaking. The combination of these three application properties necessitates poligeenan to have molecular weights in the range 10-20 kDa.
In 1969, Adrian Julian Marcus and James Watt demonstrated that poligeenan could induce gastrointestinal lesions in animal models, [6] which was followed by decades of medical research using the substance to study inflammatory bowel disease. [6]
Poligeenan is not an approved food additive in the United States. The low molecular weight of poligeenan precludes it from exhibiting functionality in food. The International Agency for Research on Cancer assigned poligeenan to carcinogenic risk category 2B (possibly carcinogenic to humans). [7] Unlike poligeenan, carrageenan is an approved food additive [8] with an extensive toxicology data base that has been reviewed by regulatory bodies and provides scientific evidence for safe inclusion in foods.
Poligeenan is produced by the harsh acid degradation of carrageenan. Carrageenan in solution is processed at low pH (~1.0) and high temperature (90 °C (190 °F)) for up to six hours until the weight-average molecular weight (Mw) has been reduced to the range 10,000 – 20,000 daltons (10–20 kDa). The resulting poligeenan solution is neutralized and spray dried to a fine powder.
Aspartame is an artificial non-saccharide sweetener 200 times sweeter than sucrose and is commonly used as a sugar substitute in foods and beverages. It is a methyl ester of the aspartic acid/phenylalanine dipeptide with brand names NutraSweet, Equal, and Canderel. Aspartame was approved by the US Food and Drug Administration (FDA) in 1974, and then again in 1981, after approval was revoked in 1980.
Guar gum, also called guaran, is a galactomannan polysaccharide extracted from guar beans that has thickening and stabilizing properties useful in food, feed, and industrial applications. The guar seeds are mechanically dehusked, hydrated, milled and screened according to application. It is typically produced as a free-flowing, off-white powder.
Sodium dodecyl sulfate (SDS) or sodium lauryl sulfate (SLS), sometimes written sodium laurilsulfate, is an organic compound with the formula CH3(CH2)11OSO3Na and structure H3C−(CH2)11−O−S(=O)2−O−Na+. It is an anionic surfactant used in many cleaning and hygiene products. This compound is the sodium salt of the 12-carbon organosulfate. Its hydrocarbon tail combined with a polar "headgroup" give the compound amphiphilic properties that make it useful as a detergent. SDS is also component of mixtures produced from inexpensive coconut and palm oils. SDS is a common component of many domestic cleaning, personal hygiene and cosmetic, pharmaceutical, and food products, as well as of industrial and commercial cleaning and product formulations.
Polyethylene glycol (PEG; ) is a polyether compound derived from petroleum with many applications, from industrial manufacturing to medicine. PEG is also known as polyethylene oxide (PEO) or polyoxyethylene (POE), depending on its molecular weight. The structure of PEG is commonly expressed as H−(O−CH2−CH2)n−OH.
Propylene glycol (IUPAC name: propane-1,2-diol) is a viscous, colorless liquid, which is nearly odorless but possesses a faintly sweet taste. Its chemical formula is CH3CH(OH)CH2OH. As it contains two alcohol groups, it is classed as a diol. It is miscible with a broad range of solvents, including water, acetone, and chloroform. In general, glycols are non-irritating and have very low volatility.
Carrageenans or carrageenins are a family of natural linear sulfated polysaccharides that are extracted from red edible seaweeds. Carrageenans are widely used in the food industry, for their gelling, thickening, and stabilizing properties. Their main application is in dairy and meat products, due to their strong binding to food proteins. In recent years, carrageenans have emerged as a promising candidate in tissue engineering and regenerative medicine applications as they resemble native glycosaminoglycans (GAGs). They have been mainly used for tissue engineering, wound coverage, and drug delivery.

Chondroitin sulfate is a sulfated glycosaminoglycan (GAG) composed of a chain of alternating sugars. It is usually found attached to proteins as part of a proteoglycan. A chondroitin chain can have over 100 individual sugars, each of which can be sulfated in variable positions and quantities. Chondroitin sulfate is an important structural component of cartilage, and provides much of its resistance to compression. Along with glucosamine, chondroitin sulfate has become a widely used dietary supplement for treatment of osteoarthritis, although large clinical trials failed to demonstrate any symptomatic benefit of chondroitin.
Barium sulfate (or sulphate) is the inorganic compound with the chemical formula BaSO4. It is a white crystalline solid that is odorless and insoluble in water. It occurs as the mineral barite, which is the main commercial source of barium and materials prepared from it. Its opaque white appearance and its high density are exploited in its main applications.

Benzyl butyl phthalate (BBP) is an organic compound historically used a plasticizer, but which has now been largely phased out due to health concerns. It is a phthalate ester of containing benzyl alcohol, and n-butanol tail groups. Like most phthalates, BBP is non-volatile and remains liquid over a wide range of temperatures. Hot-melt adhesive (HMA), also known as hot glue, is a form of thermoplastic adhesive that is commonly sold as solid cylindrical sticks of various diameters designed to be applied using a hot glue gun. The gun uses a continuous-duty heating element to melt the plastic glue, which the user pushes through the gun either with a mechanical trigger mechanism on the gun, or with direct finger pressure. The glue squeezed out of the heated nozzle is initially hot enough to burn and even blister skin. The glue is sticky when hot, and solidifies in a few seconds to one minute. Hot-melt adhesives can also be applied by dipping or spraying, and are popular with hobbyists and crafters both for affixing and as an inexpensive alternative to resin casting.

Patulin is an organic compound classified as a polyketide. It is named after the fungus from which it was isolated, Penicillium patulum. It is a white powder soluble in acidic water and in organic solvents. It is a lactone that is heat-stable, so it is not destroyed by pasteurization or thermal denaturation. However, stability following fermentation is lessened. It is a mycotoxin produced by a variety of molds, in particular, Aspergillus and Penicillium and Byssochlamys. Most commonly found in rotting apples, the amount of patulin in apple products is generally viewed as a measure of the quality of the apples used in production. In addition, patulin has been found in other foods such as grains, fruits, and vegetables. Its presence is highly regulated.
o-Toluidine (ortho-toluidine) is an organic compound with the chemical formula CH3C6H4NH2. It is the most important of the three isomeric toluidines. It is a colorless liquid although commercial samples are often yellowish. It is a precursor to the herbicides metolachlor and acetochlor.
Cobalt(II) sulfate is any of the inorganic compounds with the formula CoSO4(H2O)x. Usually cobalt sulfate refers to the hexa- or heptahydrates CoSO4.6H2O or CoSO4.7H2O, respectively. The heptahydrate is a red solid that is soluble in water and methanol. Since cobalt(II) has an odd number of electrons, its salts are paramagnetic.
2,3,7,8-Tetrachlorodibenzo-p-dioxin (TCDD) is a polychlorinated dibenzo-p-dioxin (sometimes shortened, though inaccurately, to simply 'dioxin') with the chemical formula C12H4Cl4O2. Pure TCDD is a colorless solid with no distinguishable odor at room temperature. It is usually formed as an unwanted product in burning processes of organic materials or as a side product in organic synthesis.

Sodium bisulfite (or sodium bisulphite, sodium hydrogen sulfite) is a chemical mixture with the approximate chemical formula NaHSO3. Sodium bisulfite in fact is not a real compound, but a mixture of salts that dissolve in water to give solutions composed of sodium and bisulfite ions. It appears in form of white or yellowish-white crystals with an odor of sulfur dioxide. Regardless of its ill-defined nature, sodium bisulfite is used in many different industries such as a food additive with E number E222 in the food industry, a reducing agent in the cosmetic industry, and a decomposer of residual hypochlorite used in the bleaching industry.
PhIP (2-Amino-1-methyl-6-phenylimidazo[4,5-b]pyridine) is one of the most abundant heterocyclic amines (HCAs) in cooked meat. PhIP is formed at high temperatures from the reaction between creatine or creatinine, amino acids, and sugar. PhIP formation increases with the temperature and duration of cooking and also depends on the method of cooking and the variety of meat being cooked. The U.S. Department of Health and Human Services National Toxicology Program has declared PhIP as "reasonably anticipated to be a human carcinogen". International Agency for Research on Cancer (IARC), part of World Health Organization, has classified PhIP as IARC Group 2B carcinogen. There is sufficient evidence in experimental animals, as well as in vitro models, for the carcinogenicity of PhIP.
