The party has won a seven-time majority in the Tamil Nadu Legislative Assembly and has emerged as the most successful political outfit in the state's history. It is currently the main opposition party in the Tamil Nadu Legislative Assembly.

Maruthur Gopalan Ramachandran , also popularly known by his acronym M.G.R., was an Indian actor, politician, and philanthropist who served as the Chief Minister of Tamil Nadu from 1977 until his death in 1987. He was the founder and first general secretary of the All India Anna Dravida Munnetra Kazhagam. On 19 March 1988, M.G.R. was posthumously awarded the Bharat Ratna, India's highest civilian honour.
Muthuvel Karunanidhi was an Indian writer and politician who served as Chief Minister of Tamil Nadu for almost two decades over five terms between 1969 and 2011. He is popularly referred to as Kalaignar (Artist) and Mutthamizh Arignar for his contributions to Tamil literature. He had the longest tenure as Chief Minister of Tamil Nadu with 6,863 days in office. He was also a long-standing leader of the Dravidian movement and ten-time president of the Dravida Munnetra Kazhagam political party. Karunanidhi has the record of never losing an election to the Tamil Nadu Assembly, having won 13 times since his first victory in 1957. Before entering politics, he worked in the Tamil film industry as a screenwriter. He also made contributions to Tamil literature, having written stories, plays, novels, and a multiple-volume memoir. Karunanidhi died on 7 August 2018 at Kauvery Hospital in Chennai after a series of prolonged, age-related illnesses.
Muthuvel Karunanidhi Stalin is an Indian politician serving as the 8th and current Chief Minister of Tamil Nadu. The son of the former Chief Minister M. Karunanidhi, Stalin has been the president of the Dravida Munnetra Kazhagam (DMK) party since 28 August 2018. He served as the 45th Mayor of Chennai from 1996 to 2002 and 1st Deputy Chief Minister of Tamil Nadu from 2009 to 2011. Stalin was placed 24th on the list of India's Most Powerful Personalities in 2022 by The Indian Express.

He also served as the Headquarters Secretary of the AIADMK from 2016 to 2022.
D. Jayakumar is an Indian politician and was a Member of the 15th Legislative Assembly of Tamil Nadu. He was the Speaker of the Tamil Nadu Legislative Assembly until his resignation on 29 September 2012. He was elected to the Tamil Nadu legislative assembly from Royapuram constituency as an Anna Dravida Munnetra Kazhagam candidate in the 1991, 2001, 2006, 2011 elections and 2016 elections for five terms. During the Tamil Nadu legislative Assembly 2021, he again contested in Royapuram Constituency and lost to the DMK candidate Ira Moorthy. He served in various cabinet positions starting from 1991 - Forests, Fisheries, Information Technology, Electricity and Finance. He is one among the senior leaders of AIADMK. In October 2020, he was appointed to 11 member Steering Committee of AIADMK and also serves as its Organizing Secretary. In the TN 2021 Election for Royapuram constituency, he was defeated by Murthy.R of DMK by a margin of 27,779 votes. He had represented Royapuram five times - 1991, 2001, 2006, 2011 and 2016.

The Sixteenth Tamil Nadu Legislative Assembly Election was held on 6 April 2021, to elect representatives from the 234 constituencies in the Indian state of Tamil Nadu. The Dravida Munnetra Kazhagam (DMK) won the election, ending the decade-long reign of the All India Anna Dravida Munnetra Kazhagam (AIADMK). The DMK's leader M. K. Stalin became the eighth Chief Minister of Tamil Nadu, and the 12th Chief Minister since the 1956 reorganization. He replaced Edappadi K. Palaniswami of the AIADMK.
