Winston-Salem is a city in and the county seat of Forsyth County, North Carolina, United States. In the 2020 census, the population was 249,545, making it the second-largest municipality in the Piedmont Triad region, the fifth-most populous city in North Carolina, and the 90th-most populous city in the United States. Winston-Salem is home to the tallest office building in the region, 100 North Main Street, formerly known as the Wachovia Building and now known locally as the Wells Fargo Center.
The R. J. Reynolds Tobacco Company (RJR) is an American tobacco manufacturing company based in Winston-Salem, North Carolina, and headquartered at the RJR Plaza Building. Founded by R. J. Reynolds in 1875, it is the second-largest tobacco company in the United States. The company is a wholly owned subsidiary of Reynolds American, after merging with the U.S. operations of British American Tobacco in 2004.
Wake Forest University (WFU) is a private research university in Winston-Salem, North Carolina, United States. Founded in 1834, the university received its name from its original location in Wake Forest, north of Raleigh, North Carolina. The Reynolda Campus, the university's main campus, has been located north of downtown Winston-Salem since the university moved there in 1956. The Atrium Health Wake Forest Baptist medical campus has two locations, the older one located near the Ardmore neighborhood in central Winston-Salem, and the newer campus at Wake Forest Innovation Quarter downtown. The university also occupies laboratory space at Biotech Plaza at Innovation Quarter, and at the Center for Nanotechnology and Molecular Materials. The university's Graduate School of Management maintains a presence on the main campus in Winston-Salem and in Charlotte, North Carolina.
Salem College is a private women's liberal arts college in Winston-Salem, North Carolina. Founded in 1772 as a primary school, it later became an academy and ultimately added the college. It is the oldest female educational establishment that is still a women's college and the oldest women's college in the Southern United States.

The Lawrence Joel Veterans Memorial Coliseum is a 14,665-seat multi-purpose arena, in Winston-Salem, North Carolina. Construction on the arena began on April 23, 1987, and it opened on August 28, 1989. It was named after Lawrence Joel, an Army medic from Winston-Salem who was awarded the Medal of Honor in 1967 for action in Vietnam on November 8, 1965. The memorial was designed by James Ford in New York, and includes the poem "The Fallen" engraved on an interior wall. It is home to the Wake Forest University Demon Deacons men's basketball and women's basketball teams, and is adjacent to the Carolina Classic Fairgrounds. The arena replaced the old Winston-Salem Memorial Coliseum, which was torn down for the LJVM Coliseum's construction.

Innovation Quarter in Winston-Salem, North Carolina, formerly Wake Forest Innovation Quarter, is an innovation district focused on research, business, and education in biomedical science, information technology, digital media, clinical services, and advanced materials. The Innovation Quarter, operated by Wake Forest Baptist Medical Center, is home to academic groups, private companies and other organizations located on 330 acres in downtown Winston-Salem. Its tenants include departments from five academic institutions—Wake Forest School of Medicine, Wake Forest University, Forsyth Technical Community College, Winston-Salem State University, UNC School of the Arts— as well as private businesses and other organizations. One tenant is the Wake Forest Institute for Regenerative Medicine (WFIRM), which is working to engineer more than 30 different replacement tissues and organs and to develop healing cell therapies. The science and research conducted at WFIRM is behind two start-up companies at Innovation Quarter. The ability of researchers and scientists to work alongside entrepreneurs furthers a goal of Innovation Quarter to develop new treatments and cures for disease and advances in technology.

It is affiliated with Atrium Health Wake Forest Baptist, the academic medical center whose clinical arm is Atrium Health Wake Forest Baptist. In 2021, U.S. News & World Report ranked Wake Forest School of Medicine 48th best for research in the nation and 80th best for primary care. The School of Medicine also ranks in the top third of U.S. medical schools in funding from the National Institutes of Health (NIH).

Allegacy Federal Credit Union Stadium is a football stadium in Winston-Salem, North Carolina. The stadium is just west of Gene Hooks Field at Wake Forest Baseball Park, home of the Wake Forest baseball team. It is primarily used for American football, and is the home field of the Wake Forest University Demon Deacons. The stadium opened in 1968 and holds 31,500 people. It is the smallest football stadium, by permanent capacity, in both the ACC and in all Power Five conferences. Previously known as Groves Stadium, in September 2007, Wake Forest University and BB&T, which was headquartered in Winston-Salem, announced a 10-year deal to officially rename the stadium BB&T Field starting with the first 2007 home game against Nebraska. The deal was part of a larger development process to secure funds for stadium renovations and upgrades. On July 8, 2020, the name of the stadium was changed to Truist Field at Wake Forest following a merger between BB&T and SunTrust. On June 21, 2023, the stadium name was changed to Allegacy Federal Credit Union Stadium after Allegacy became an official banking partner with Wake Forest Athletics.

The other church was on the campus of Wake Forest University in Winston-Salem, North Carolina. It was active from 1956 to 2022, but has now closed.

Mary Reynolds Babcock was an American philanthropist. As the daughter of R.J Reynolds and Katharine Smith Reynolds, she therefore inherited considerable wealth from her father's company, the nationally prominent R. J. Reynolds Tobacco Company. She was a founder for both the Z. Smith Reynolds Foundation and the Mary Reynolds Babcock Foundation. O'Kelly Library at Winston-Salem State University and a leader in state and national library associations. She is the first American Library Association president who is a library director at one of the nation's historically black colleges and universities.

Smith Reynolds Library at Wake Forest University in Winston-Salem, North Carolina. She has been invited to speak on several occasions as an expert historian and archivist across the United States.
Reynolda Hall is the main building on the Reynolda Campus of Wake Forest University in Winston-Salem, North Carolina. It is located on the southeastern side of Hearn Plaza, opposite Wait Chapel, while the rear of the building overlooks Manchester Plaza.
