Mosima Gabriel "Tokyo" Sexwale is a South African businessman, politician, anti-apartheid activist, and former political prisoner. Sexwale was imprisoned on Robben Island for his anti-apartheid activities, alongside figures such as Nelson Mandela. After the 1994 general election—the first fully democratic election in South Africa—Sexwale became the Premier of Gauteng Province. He served in the government of South Africa as Minister of Human Settlements from 2009 to 2013.
Nkosazana Clarice Dlamini-Zuma, sometimes referred to by her initials NDZ, is a South African politician, doctor and anti-apartheid activist, currently serving as Minister of Cooperative Governance and Traditional Affairs. She was South Africa's Minister of Health from 1994 to 1999, under President Nelson Mandela, Minister of Foreign Affairs, under both President Thabo Mbeki and President Kgalema Motlanthe, Minister of Home Affairs in the first term of former President Jacob Zuma and Minister in the Presidency for the National Planning Commission for Policy and Evaluation under President Cyril Ramaphosa.

Elizabeth Dipuo Peters was the Minister of Transport of the Republic of South Africa from 10 July 2013 until 30 March 2017, in the Zuma administration, and former Minister of Energy from 2009 to 2013 having served as successor to Manne Dipico as the second Premier of the Northern Cape Province, 22 April 2004 to 10 May 2009. A member of the African National Congress (ANC), she serves on the Women's League National Executive Committee. Dipuo Peters resigned as a member of parliament for the African National Congress in April 2017.
The Department of Human Settlements is the department under the Ministry of Human Settlements, of the South African government responsible for housing and urban development. Its primary purpose is the implementation of the constitutional mandate that "everyone has the right to have access to adequate housing." It works in co-operation with the provincial governments, each of which has its own Human Settlements department, and with the municipalities.

Along with Michael Masutha she is one of only two leaders with disability in the current regime. She is a motivational speaker, HIV & AIDS activist, business woman and co-founded Disabled Youth South Africa. She was instrumental in drafting the country’s UN Convention on the Rights of Persons with Disabilities’ Country Report.

The Second Cabinet of Cyril Ramaphosa was formed on 29 May 2019 after President Ramaphosa was inaugurated for his first full-term as President of South Africa following the African National Congress's victory in the 2019 South African general election. The cabinet was slightly reduced from 36 portfolios to only 28 portfolios due to the amalgamation of several departments. The cabinet is the third cabinet in Africa to be made completely gender-equal in its composition and the first gender-equal cabinet in South African history.
