Lieber Institute for Brain Development

The Lieber Institute for Brain Development (LIBD) is a nonprofit research center located in Baltimore, Maryland, that studies brain development issues such as schizophrenia and autism. [1] The cause of most neuropsychiatric disorders remains unknown and current therapies such as antipsychotics and antidepressants treat symptoms rather than the underlying illness. Lieber is working to unravel the biological basis of these brain disorders and is developing therapies to treat or prevent their development. [2]

History and description

The Lieber Institute was started in 2010 by Steve and Connie Lieber along with Milton and Tamar Maltz—both families have children with schizophrenia. [3] [4] [5] Psychiatrist Daniel R. Weinberger is the CEO. [6] Having gathered over 3,000 brains for research purposes, the institute has the largest collection of post-mortem brains diagnosed with post-traumatic stress disorder. [7] [2] These brains are used to study the biological basis for mental illnesses. [8] In one study of post-mortem brains, LIBD researchers found that patients with schizophrenia had high levels of DNA methylation in brain regions associated with risk for schizophrenia. [9] They are also developing a drug to prevent post-traumatic stress disorder. [10]

Research and partnerships

In 2011, researchers who later joined the Lieber Institute published a review of brain banks for studying diseases such as schizophrenia, alzheimer’s diseases, bipolar disorder, and other psychiatric illnesses. Postmortem human brain tissue is critical for advancing neurobiological studies of psychiatric illness. [11]

In a December 2015 paper in Neuron, the Lieber Institute announced the formation of a new consortium called BrainSeq, along with seven pharmaceutical partners. The consortium’s goal is to analyze post mortem brain samples for DNA, RNA and DNA-methylation differences establish a public database. [12]

A paper by Lieber scientists found that altering brain proteins caused the cells of autistic brains to behave normally. [13] The specific type of autism is called Pitt–Hopkins syndrome and researchers are looking into possible treatments that could be used for the condition and for other types of autism. [14] [15]

Researchers with Lieber published a paper in Nature Medicine that examined the placenta and found that genes and pregnancy problems combine to increase the likelihood of developing schizophrenia. [16] [17] This study adds to growing evidence that the placenta is critical to understanding the health and course of pregnancy. [18]

African Ancestry Neuroscience Research Initiative

In 2019, the Lieber Institute announced a new initiative to study brain diseases in African Americans. The venture is a partnership with the African-American Clergy Medical Research Initiative, a group of clergy leaders in Baltimore. Research suggests that neuropsychiatric diagnoses are 20% more frequent in African-American communities than in communities of European ancestry. [19] [20] In studies of brain disorders, individuals of African descent make up, on average, less than 5% of research cohorts. Lieber Institute Director Daniel R. Weinberger wrote that individuals of African ancestry must be included in research for brain illnesses. [21]

The African Ancestry Neuroscience Research Initiative will generate brain gene and protein expression profiles for people of African ancestry, a group that is greatly underrepresented in research examining the genetic causes of diseases. Lack of research involving African Americans is problematic because African Americans are 20% more likely to experience serious mental health problems than the general population and are about twice as likely to develop Alzheimer’s disease. Suicide rates for African American children aged 5–11 are twice as high as comparable individuals of European ancestry even after controlling for socioeconomic factors. [22] In the summer of 2020, Governor Larry Hogan of Maryland budgeted $1.25 million to the effort, with another $1 million coming from Brown Capital, one of the oldest African-American founded asset management firms in the country. [23]

Institute of Psychiatry, Psychology and Neuroscience

The Institute of Psychiatry, Psychology and Neuroscience (IoPPN) is a research institution dedicated to discovering what causes mental illness and diseases of the brain. In addition, its aim is to help identify new treatments for them and ways to prevent them in the first place. The IoPPN is a faculty of King's College London, England, previously known as the Institute of Psychiatry (IoP).

Neuropsychiatry or Organic Psychiatry is a branch of medicine that deals with psychiatry as it relates to neurology, in an effort to understand and attribute behavior to the interaction of neurobiology and social psychology factors. The biopsychiatry controversy is a dispute over which viewpoint should predominate and form a basis of psychiatric theory and practice. The debate is a criticism of a claimed strict biological view of psychiatric thinking. Its critics include disparate groups such as the antipsychiatry movement and some academics.

Psychiatric genetics is a subfield of behavioral neurogenetics and behavioral genetics which studies the role of genetics in the development of mental disorders. The basic principle behind psychiatric genetics is that genetic polymorphisms are part of the causation of psychiatric disorders.

DISC1 Protein-coding gene in the species Homo sapiens

Disrupted in schizophrenia 1 is a protein that in humans is encoded by the DISC1 gene. In coordination with a wide array of interacting partners, DISC1 has been shown to participate in the regulation of cell proliferation, differentiation, migration, neuronal axon and dendrite outgrowth, mitochondrial transport, fission and/or fusion, and cell-to-cell adhesion. Several studies have shown that unregulated expression or altered protein structure of DISC1 may predispose individuals to the development of schizophrenia, clinical depression, bipolar disorder, and other psychiatric conditions. The cellular functions that are disrupted by permutations in DISC1, which lead to the development of these disorders, have yet to be clearly defined and are the subject of current ongoing research. Although, recent genetic studies of large schizophrenia cohorts have failed to implicate DISC1 as a risk gene at the gene level, the DISC1 interactome gene set was associated with schizophrenia, showing evidence from genome-wide association studies of the role of DISC1 and interacting partners in schizophrenia susceptibility.

Neurogenomics

Neurogenomics is the study of how the genome of an organism influences the development and function of its nervous system. This field intends to unite functional genomics and neurobiology in order to understand the nervous system as a whole from a genomic perspective.

Social constructionism, a branch of sociology, queries commonly held views on the nature of reality, touching on themes of normality and abnormality within the context of power and oppression in societal structures.

The Semel Institute for Neuroscience and Human Behavior is a research institute of the University of California Los Angeles (UCLA). It includes a number of centers, including the "Center for Neurobehavioral Genetics", which uses DNA sequencing, gene expression studies, bioinformatics, and the genetic manipulation of model organisms to understand brain and behavioral phenotypes.

Childhood schizophrenia is similar in characteristics of schizophrenia that develops at a later age, but has an onset before the age of 13 years, and is more difficult to diagnose. Schizophrenia is characterized by positive symptoms that can include hallucinations, delusions, and disorganized speech; negative symptoms, such as blunted affect and avolition and apathy, and a number of cognitive impairments. Differential diagnosis is problematic since several other neurodevelopmental disorders, including autism spectrum disorder, language disorder, and attention deficit hyperactivity disorder, also have signs and symptoms similar to childhood-onset schizophrenia.

Perminder Sachdev is a neuropsychiatrist based in Australia. He is the Scientia Professor of Neuropsychiatry at the University of New South Wales (UNSW), Australia, the director of the Centre for Healthy Brain Ageing (CHeBA), UNSW, and the Clinical Director of the Neuropsychiatric Institute (NPI) at the Prince of Wales Hospital, Sydney, Australia.

Cognitive genomics is the sub-field of genomics pertaining to cognitive function in which the genes and non-coding sequences of an organism's genome related to the health and activity of the brain are studied. By applying comparative genomics, the genomes of multiple species are compared in order to identify genetic and phenotypical differences between species. Observed phenotypical characteristics related to the neurological function include behavior, personality, neuroanatomy, and neuropathology. The theory behind cognitive genomics is based on elements of genetics, evolutionary biology, molecular biology, cognitive psychology, behavioral psychology, and neurophysiology.

Lori L. Altshuler American psychiatrist

Lori Altshuler was a Professor at the University of California, Los Angeles (UCLA) Department of Psychiatry and Biobehavioral Sciences and held the Julia S. Gouw Endowed Chair for Mood Disorders. Altshuler was the Director of the UCLA Mood Disorders Research Program and the UCLA Women's Life Center, each being part of the Neuropsychiatric Hospital at UCLA.

Daniel R. Weinberger is a professor of psychiatry, neurology and neuroscience at Johns Hopkins University and Director and CEO of the Lieber Institute for Brain Development, which opened in 2011.

Carol A. Tamminga is an American psychiatrist and neuroscientist, focusing in treating psychotic illnesses, such as schizophrenia, psychotic bipolar disorder, and schizoaffective disorder, currently the Lou and Ellen McGinley Distinguished Chair in Psychiatric Research and the Chief of the Translational Neuroscience Division in Schizophrenia at University of Texas Southwestern Medical Center. She hss been Chair of the Department of Psychiatry at UTSW since 2008. She is an Elected Fellow of the National Academy of Medicine. She serves on the advisory boards of the Brain and Behavioral Research Foundation and of the National Institute of Mental Health (NIMH). In 2011 she was awarded the Lieber Prize for Outstanding Achievement in Schizophrenia Research. Tamminga led a study examining whether giving Prozac to fetuses with Down syndrome would improve the functioning of their brains. In her attempt to confirm psychiatric diagnoses biologically, she found "biotypes" or "clusters." Her current research involves mechanisms underlying schizophrenia, especially its most prominent symptoms, psychosis and memory dysfunction.

Sergiu P. Pașca Romanian-American scientist and [[physician]] at [[Stanford University]]

Sergiu P. Pașca is a Romanian-American scientist and physician at Stanford University in California. Pașca is a faculty member in the Department of Psychiatry and Behavioral Sciences at Stanford University and the Uytengsu Family Director of Stanford Brain Organogenesis, a neuroscientist and stem cell biologist and currently a NYSCF Robertson Investigator. He is part of the Stanford Neurosciences Institute, Stanford Bio-X and a fellow of the ChEM-H Institute at Stanford. Pașca was listed among New York Times Visionaries in Medicine and Sciences, and he is the recipient of the 2018 Vilcek Award for Creative Biomedical Promise from the Vlicek Foundation.

Ming Tso Tsuang is an American psychiatrist and Distinguished Professor of Psychiatry at the University of California, San Diego. He is considered a pioneering researcher in the genetic epidemiology of schizophrenia and other severe mental disorders. Tsuang has authored and co-authored more than 600 publications and serves as founding and senior editor of the American Journal of Medical Genetics Part B.

The PsychENCODE Consortium was founded by the U.S. National Institutes of Health in 2015 to study the role of rare genetic variants involved in several psychiatric disorders. PsychENCODE aims to create a public resource of genomic data gathered from 1,000 healthy and disease-affected post-mortem brains reflecting different developmental periods.

Joseph T. Coyle

Joseph Thomas Coyle Jr. born in Chicago, Illinois, is a psychiatrist and neuroscientist that is known for his work on the neurobiology of mental illness, more specifically on schizophrenia. He is currently the Eben S. Draper Chair of Psychiatry and Neuroscience at Harvard Medical School in Boston, Massachusetts. He was President of the Society of Neuroscience from 1991–1992. [2] and also the president of the American College of Neuropsychopharmacology in 2001, He is a member of the Institute of Medicine.
