The 2021 LTP Charleston Pro Tennis II was a professional women's tennis tournament played on outdoor clay courts. It was the sixth edition of the tournament which was part of the 2021 ITF Women's World Tennis Tour. It took place in Charleston, South Carolina, United States between 21 and 27 June 2021.

This was the first edition of the tournament.

Shelby Rogers was the defending champion, but chose not to participate.

Caroline Dolehide and Maria Sanchez were the defending champions but chose not to participate.

Cara Black and Arina Rodionova were the defending champions, having won the previous edition in 2012. However Black retired from professional tennis in 2015 and Rodionova chose to compete at the 2022 Melbourne Summer Set instead.

Paige Hourigan and Astra Sharma were the defending champions having won the previous edition in 2019, however both players chose not to participate.

Astra Sharma and Aldila Sutjiadi defeated Emina Bektas and Tara Moore in the final, 4–6, 6–4, [11–9], to win the doubles tennis title at the 2022 Copa Colsanitas. The win earned Sharma her second Copa Colsanitas doubles title after first winning in 2019, and Sutjiadi her first career Women's Tennis Association (WTA) title.
