The channel carries comedy programming in the form of both original, licensed, and syndicated series, stand-up comedy specials, and feature films.
Jackhole Productions is an American production company started by Jimmy Kimmel, Daniel Kellison and Adam Carolla. It has produced several comedy shows on television. Jackhole Productions has worked on several projects with production company DiGa. The company's name is an amalgam of Carolla and Kellison's company Jackhouse, and Kimmel's company Cashhole. It is also a made up swear word. The mascot is an animated donkey wearing a sombrero. In early 2020, Kimmel's new production company, Kimmelot, assumed production responsibilities for Jimmy Kimmel Live!; it is not known if this is due to a merger between Jackhole and Kimmelot.
The Ben Stiller Show is an American sketch comedy series starring Ben Stiller that aired in two iterations, the first series airing six episodes on MTV in 1990, and the second series airing 12 episodes on Fox from September 27, 1992, to January 17, 1993, with a 13th episode airing in 1995 on Comedy Central. The Fox version also starred Andy Dick, Janeane Garofalo and Bob Odenkirk. Character actor John F. O'Donohue also appeared in every episode.

David Benjamin Wain is an American comedian, writer, actor, and director. He has co-written and directed six feature films, including Wet Hot American Summer (2001), Role Models (2008), Wanderlust (2012) and They Came Together (2014). He has also served as a creator, producer, writer and director on a number of television series, including Wet Hot American Summer: Ten Years Later, Wet Hot American Summer: First Day of Camp, Childrens Hospital and Medical Police. He has had small roles in most of the films and TV series he has produced or directed. Wain had a starring voice role as The Warden on the 2008–2014 Adult Swim animated series Superjail!, and has provided the voices for Courtney Wheeler and Grover Fischoeder on Bob's Burgers since 2012.

Saul of the Mole Men is an American live-action/puppet/animated hybrid comedy television series created by Craig Lewis, former writer on Cartoon Network's The Grim Adventures of Billy & Mandy and Foster's Home for Imaginary Friends. The series first aired on the channel's late night Adult Swim programming block on February 11, 2007. Described as "an ultra-patriotic Land of the Lost set in the center of the Earth", the series was directed by Tom Stern and stars Josh Gardner, who previously collaborated with Stern on the television series Gerhard Reinke's Wanderlust. Its theme song is performed by South Park co-creator Trey Parker.
Josh Gardner is an American comedian and writer, Gardner is best known for his role as Saul Malone, a Gee-al-agist, on Adult Swim's cult-classic Saul of the Mole Men. Gardner first appeared on TV under the name Gerhard Reinke, the German host of the Comedy Central travel show, Gerhard Reinke's Wanderlust, in 2003. Gardner also wrote for Comedy Central's The Man Show, and the Fox series A Minute With Stan Hooper. Aside from comedy, Gardner wrote for television game shows such as You Don't Know Jack and Sports Geniuses. He was also a regular contributor as the character Deaf Frat Guy on The Adam Carolla Show, and continues to appear on The Adam Carolla Podcast. Josh is a graduate of Salisbury School and Hamilton College. On April 6, 2011 Josh released his album of original songs on iTunes titled Mr. Stinkfinger. On the February 22nd episode of the Adam Carolla Podcast, Josh announced his forthcoming album "Winterbush", which will feature a much more "wintery" feel after the "autumnal" vibe of his last album. Nat Faxon is Gardner's cousin.

Stella is a comedy trio or comedy troupe consisting of Michael Showalter, Michael Ian Black, and David Wain. The group formed in 1997 as a weekly nightclub comedy attraction, performing at New York City nightclub Fez from 1997 until the club's closing in February 2005. Stella soon gained a wider cult following after a series of self-produced shorts known as the Stella shorts were released in limited quantities on DVD beginning in 2002. The trio later created and starred in a Stella television series in 2005. Now known for their unique blend of potentially mainstream comedy and surrealist humor, Stella has garnered a small but dedicated fanbase.
Comedy Central Roast is a series of celebrity roast specials that air on the American television channel Comedy Central. The first official Comedy Central Roast premiered on August 10, 2003. On average one or two roasts air every year. There are eight to ten people invited who roast each other before finally roasting the title subject of each show. As of 2019, seventeen roasts have aired. Targets of roasts have included musicians, actors and comedians. Since 2010, Comedy Central affiliates outside the United States have occasionally produced their own roasts; twelve such roasts have aired so far, in five countries.
