Sexual maturation disorder is a disorder of anxiety or depression related to an uncertainty about one's gender identity or sexual orientation. The World Health Organization (WHO) lists sexual maturation disorder in the ICD-10, under "Psychological and behavioural disorders associated with sexual development and orientation". In the ICD-10 it is explicitly stated that sexual orientation, by itself, is not a disorder and is not classified under this heading. Sexual maturation disorder, along with ego-dystonic sexual orientation and sexual relationship disorder, was introduced to the ICD in 1990, replacing the ICD-9 diagnosis of homosexuality. The diagnosis is not included in the ICD-11.

Puberty blockers, also called puberty inhibitors, are drugs used to postpone puberty in children. The most commonly used puberty blockers are gonadotropin-releasing hormone (GnRH) agonists, which inhibit the release of sex hormones, including testosterone and estrogen. In addition to their various other medical uses, puberty blockers are used off-label for transgender children to delay the development of unwanted sex characteristics, with the intent to provide transgender youth more time to explore their identity. The use of puberty blockers in transgender youth has been challenged on ethical and medical grounds, causing controversy over the morality and legality of their use.

The Center of Expertise on Gender Dysphoria (CEGD), or the Kennis- en Zorgcentrum Genderdysforie (KZcG), is a transgender clinic at the VU University Medical Center (VUMC) of the Vrije Universiteit Amsterdam in Amsterdam, Netherlands. It opened in 1972 and is one of the largest transgender clinics and research institutes in the world. As of 2021, it has treated about 10,000 transgender people since it opened almost 50 years previously. The clinic was first headed by Louis Gooren.

The European Network for the Investigation of Gender Incongruence (ENIGI) is a collaborative multicenter prospective cohort study on gender incongruence which started in 2010 and is being conducted by several transgender clinics in Europe. The clinics that have been involved in the initiative include the Center of Expertise on Gender Dysphoria (CEGD) at the VU University Medical Center (VUMC) in Amsterdam, Netherlands, the Center for Sexology and Gender at Ghent University Hospital in Ghent, Belgium, the University Medical Center Hamburg-Eppendorf in Hamburg, Germany, the Oslo University Hospital, Rikshospitalet in Oslo, Norway, and the University Hospital of the University of Florence in Florence, Italy. The clinics in the ENIGI initiative developed a common study and treatment protocol and maintain a shared database. The study includes an endocrine part to evaluate the effects of transgender hormone therapy in transfeminine and transmasculine people.
