A total of 308 players and 30 coaches,including Pakistani and Foreigners were divided into 5 different categories Platinum,Diamond,Gold,Silver and Emerging. Each franchise had a salary-spending cap of $1.2 million,including the signing of players,coaches and support staff. There were five icon players and each team was allowed to pick one icon and six foreigners in a 20-man squad.

This was supposed to be the second season held entirely in Pakistan. It was scheduled initially to be held in February and March 2021.
The players draft for the 2020 Pakistan Super League took place at National Cricket Academy,Lahore on 6 December 2019. The first round took place on 3 November 2019 to determine pick order. As this season was scheduled to be held entirely in Pakistan,only the players who were comfortable with playing in Pakistan were included in this draft.

The Karachi–Lahore cricket rivalry,known in PSL context as Karachi Kings–Lahore Qalandars rivalry,is an oldest city rivalry between cricket teams Karachi and the Lahore. It is considered to be the biggest rivalry in the PSL due to the size of the cities. The rivalry has been fuelled by the size of the two cities that they play for and their historic economic and cultural rivalry. A Karachi–Lahore clash is known typically as a heated contest which "brings out the best in players". The two cities share a rivalry in domestic cricket that dates back to the 1950s and 1960s,with the PSL providing a high-voltage platform that allows it to thrive. It is sometimes also known as the El-Clasico of PSL.
The players draft for the sixth edition of the 2021 Pakistan Super League,which is the franchise T20 cricket league of Pakistan,took place on 10 January 2021. Before the draft,the teams were allowed to retain a maximum of 8 players and make any transfers that they saw necessary. More than 400 players from over 20 different countries featured at the draft. From these players,the teams each selected 16 players along with 2 extra supplementary players to complete their 18 man squads. The draft was completed one month before the main tournament the 2021 Pakistan Super League started,in late February. Due to the suspension of the tournament because of COVID-19,a replacement draft was held.
