Thirteenth Assembly of Tamil Nadu was instituted after the victory of DMK and allies, in the 2006 state assembly election. M. Karunanidhi became the 14th Chief Minister of Tamil Nadu due to the election.

K. Ponmudi is an Indian politician and the current Minister for Higher Education of Tamil Nadu. He was the Minister for Higher Education of Tamil Nadu from 2006 to 2011. He was born in T. Edaiyar in the Villupuram district. He has a bachelor's degree in law, a master's degree in history, political science and public Administration and a doctorate in political science. He is an alumnus of Annamalai University. Before entering professional politics, he was a professor in the Villupuram government college. He has been elected to the Tamil Nadu assembly five times. From 1989 to 1991 during Dravida Munnetra Kazhagam rule he was the Minister for Health, and from 1996 to 2001 he was the Minister for Transport and Highways. He authored a book titled Dravidian Movement in India and Black Movement in the U.S.A.

E. V. Velu is an Indian and Minister of Public works, Highways and Minor Ports for Tamilnadu. He served as former Minister for Food in the state of Tamil Nadu, India. He was praised as " Ethilum vallavar Velu" by former chief minister Kalaignar Karunanithi.

Thangam Thenarasu is an Indian politician and the Minister for Finance, Planning, Human Resources Management, Pensions and Pensionary benefits, Statistics and Archeology Minister of Tamil nadu. He was allocated Electricity and Non-Conventional Energy Development portfolios of minister V. Senthil Balaji who was arrested by the Enforcement Directorate in a job racket case on 16 June 2023 by Tamil Nadu Governor R. N. Ravi on the recommendations of Chief Minister M K Stalin.

The second legislative assembly election to the Madras state was held on 31 March 1957. This was the first election held after the linguistic reorganization of Madras state in 1954. Indian National Congress and its leader K. Kamaraj won the election, and defeated its rival Dravida Munnetra Kazhagam. In 1954, due to the resignation of C. Rajagopalachari, for his controversial Kula Kalvi Thittam, the leadership of Congress was contested, between K. Kamaraj, and C. Subramaniam. Eventually, K. Kamaraj, won the support of the party, was elected leader and chief minister of Madras State in 1954. In a surprise move, he appointed both M. Bhaktavatsalam and C. Subramaniam, in his cabinet, allowing great unity amongst Congress, that ruled the state of Madras, for the next decade. This election saw future DMK leaders, M. Karunanidhi and K. Anbazhagan winning their first MLA seat in the legislative assembly.

Edappadi Karuppa Palaniswami, often referred to by his initials E.P.S., is an Indian politician who is the former chief minister of Tamil Nadu and the current leader of opposition in the Tamil Nadu Legislative Assembly. He served as the 7th chief minister of Tamil Nadu, from 2017 to 2021. He has been the General Secretary of All India Anna Dravida Munnetra Kazhagam since 28 March 2023. Palaniswami served as the Joint Coordinator of the AIADMK from 2017 to 2022 and Interim General Secretary of the AIADMK from 2022 to 2023. He also served as the Headquarters Secretary of the AIADMK from 2016 to 2022.

J. Jayalalitha assumed office as Chief Minister of Tamil Nadu for the fourth time.
