DNA polymerase epsilon subunit 3 is an enzyme that in humans is encoded by the POLE3 gene. [5] [6] [7]

Contents

POLE3 is a histone-fold protein that interacts with other histone-fold proteins to bind DNA in a sequence-independent manner. These histone-fold protein dimers combine within larger enzymatic complexes for DNA transcription, replication, and packaging.[supplied by OMIM] [7]

Interactions

POLE3 has been shown to interact with SMARCA5. [6]

Histone H2A.Z is a protein that in humans is encoded by the H2AZ1 gene.

Histones are basic nuclear proteins that are responsible for the nucleosome structure of the chromosomal fiber in eukaryotes. Nucleosomes consist of approximately 146 bp of DNA wrapped around a histone octamer composed of pairs of each of the four core histones. The chromatin fiber is further compacted through the interaction of a linker histone, H1, with the DNA between the nucleosomes to form higher order chromatin structures. The H2AFZ gene encodes a replication-independent member of the histone H2A family that is distinct from other members of the family. Studies in mice have shown that this particular histone is required for embryonic development and indicate that lack of functional histone H2A leads to embryonic lethality.

Elongator complex protein 3, also named KAT9, is a protein that in humans is encoded by the ELP3 gene. ELP3 is the catalytic histone acetyltransferase subunit of the RNA polymerase II elongator complex, which is a component of the RNA polymerase II holoenzyme and is involved in transcriptional elongation. ELP3 supports the migration and branching of projection neurons through acetylation of alpha-tubulin in the developing cerebral cortex. In mammals, ELP3 is important for paternal DNA demethylation after fertilization. ELP3 is potentially involved in cellular redox homeostasis by mediating the acetylation of glucose-6-phosphate dehydrogenase. Besides, ELP3 may play a role in chromatin remodeling and is involved in acetylation of histones H3 and probably H4.
