The 2016 Istanbul Open was a men's tennis tournament played on outdoor clay courts. It was the second edition of the Istanbul Open, and an ATP World Tour 250 event. It took place at the Koza World of Sports Arena in Istanbul, Turkey, from 25 April – 1 May 2016.

Rafael Nadal defeated the two-time defending champion Kei Nishikori in the final, 6–4, 7–5 to win the singles title at the 2016 Barcelona Open. It was Nadal's record-extending ninth title at the Barcelona Open and his 49th clay court title, equaling Guillermo Vilas' record for the most titles on clay.

Cameron Norrie defeated Nikoloz Basilashvili in the final, 3–6, 6–4, 6–1 to win the men's singles tennis title at the 2021 Indian Wells Masters. It was his first ATP Tour Masters 1000 title, and he became the first Briton to win the title. Basilashvili became the first Georgian to reach a Masters final since Alex Metreveli in 1968, and the first player to do so representing Georgia as an independent nation.
