Alex Jan"Lex"van der Eb (born 16 January 1934) is a Dutch molecular biologist and virologist. He was a professor of fundamental tumor virology and later molecular carcinogenesis at Leiden University from 1979 to 2000. He has performed research in adenoviruses and was fundamental in the creation of the technique of calcium phosphate transfection and the founding of the HEK 293 and PER.C6 cell lines.
Van der Eb was born on 16 January 1934 in Bandung,Dutch East Indies. [1] He studied biology at Leiden University and afterwards attended Delft University of Technology for a further year to specialize in microbiology. [2] Van der Eb had to fulfill his military service upon graduation and asked the head of military health services to be employed in one of the military laboratories. In 1962 he was sent to work as a biologist on poxviruses in the virology department of Leiden University. At Leiden he met professor J.A. Cohen of the Medical-Biological Laboratory of the Netherlands Organisation for Applied Scientific Research (TNO). After his military service ended in 1963 van der Eb became a grad-student under Cohen. [2] [3] He studied adenoviruses and characterized the virus DNA in 1966. [4] He obtained his PhD at Leiden University in 1968 with a thesis titled:"Fysisch-chemische en biologische eigenschappen van het DNA van dierlijke tumorvirussen". [1] Van der Eb subsequently moved to the United States to become a post-grad at the California Institute of Technology,where he broadened his knowledge of DNA. [2]
Upon request of Cohen van der Eb returned to the Netherlands in 1970 to start a research group in molecular biology of oncoviruses. [2] During this period Canadian post-doc Frank L. Graham joined his research group. [4] Together they worked on a technique to insert foreign DNA in cultured human cells. The technique of calcium phosphate transfection became available in 1973. [5] [6] [7] Fellow molecular biologist Piet Borst called this discovery of how to insert foreign DNA in human cells the cornerstone of genetics and cell biology. [3] [8] In 1972 van der Eb obtained and managed to culture embryonic kidney cells from an aborted or miscarried human foetus. [9] [10] With Graham's contributions in transfection of the cells with the adenovirus 5 this led to the cell line of HEK 293 cells. [4] [11] HEK 293 became one of the most used cell lines worldwide,coming close to the use of the HeLa cell line. [9] In 1974 van der Eb and Graham demonstrated that loose virus DNA could cause cancer. [5]
In 1974 van der Eb became a lector of fundamental tumor virology. In 1979 he was named professor of fundamental tumor virology. [1] From 1987 to 1992 he performed research on the chicken anemia virus together with Mathieu Noteborn. [12] In 1988 his teaching assignment was changed to molecular carcinogenesis. [1] In 1995,together with Dinko Valerio,he created the PER.C6 cell line with enhanced traceability compared to the HEK 293 line,this time coming from retinal cells. [4] The goal of Valerio was to use it for gene therapy but this was ultimately unsuccessful. In contrast to HEK 293 the PER.C6 cell line was patented by Leiden University and IntroGene (later Crucell) and used commercially. [3] [9] AdVac later became an enhanced version of PER.C6 and both were deemed to be very useful to construct vaccines with,becoming the basis of vaccines for ebola,Zika fever and HIV. [3] Van der Eb retired in 2000. [1] He later worked as an advisor for biotechnology company Crucell between 2000 and 2013. [3] In 2007 he was involved in writing the behavioural code for biosecurity of the Royal Netherlands Academy of Arts and Sciences. [13]
In 2020 van der Eb together with two other former Crucell officials stated that the Netherlands would have had a better starting position in obtaining vaccines to deal with the COVID-19 pandemic in the Netherlands if the government had been interested in letting Crucell work together with the Netherlands Vaccine Institute ,with Crucell offering the Netherlands a rapid vaccine production in the case of a pandemic multiple times during the early 2000s. Crucell was later bought by the American corporation Johnson &Johnson. [3]
Van der Eb was elected a member of the European Molecular Biology Organization in 1977. [14] In 1978 he won the M.W. Beijerinck Virology Medal of the Royal Netherlands Academy of Arts and Sciences. [15] He was elected member of the Royal Netherlands Academy of Arts and Sciences in 1987. [16] He was elected a member of Academia Europaea in 1989. [17] In 1989 he was also winner of the Robert Koch Prize. [18] In the same year he won the Japan Foundation for Promotion of Cancer Research Award. [19] He was made a Knight of the Order of the Netherlands Lion in 1998. [20] In 2003 he was appointed an honorary member of the Netherlands Society of Gene and Cell Therapy. [21] He is a member of the Koninklijke Hollandsche Maatschappij der Wetenschappen. [3]
Transfection is the process of deliberately introducing naked or purified nucleic acids into eukaryotic cells. It may also refer to other methods and cell types,although other terms are often preferred:"transformation" is typically used to describe non-viral DNA transfer in bacteria and non-animal eukaryotic cells,including plant cells. In animal cells,transfection is the preferred term as transformation is also used to refer to progression to a cancerous state (carcinogenesis) in these cells. Transduction is often used to describe virus-mediated gene transfer into eukaryotic cells.
Human embryonic kidney 293 cells,also often referred to as HEK 293,HEK-293,293 cells,or less precisely as HEK cells,are a specific immortalised cell line derived from a spontaneously miscarried or aborted fetus or human embryonic kidney cells grown in tissue culture taken from a female fetus in 1973.
Regnier de Graaf,original Dutch spelling Reinier de Graaf,or Latinized Reijnerus de Graeff,was a Dutch physician,physiologist and anatomist who made key discoveries in reproductive biology. He specialized in iatrochemistry and iatrogenesis,and was the first to develop a syringe to inject dye into human reproductive organs so that he could understand their structure and function.
Elie Aron Cohen was a Dutch medical doctor who,being Jewish,was sent to the Auschwitz concentration camp. He arrived there on 16 September 1943. His first wife,his first son as well as his parents-in-law were killed upon arrival,but he managed to survive through a combination of chance and skill. His status and abilities as a doctor were instrumental for his survival. On 6 May 1945 he was liberated by the U.S. military in Melk (Austria),where he had been transported by way of Mauthausen-Gusen. After World War II,Elie Cohen remarried a Jewish woman. They have two children,a daughter and a son. Elie Cohen is the author of a number of books about the Holocaust. The first of these was the Ph.D. thesis on which he graduated on 11 March 1952,at Utrecht State University. The book was entitled "The German Concentration Camp —a medical and psychological study",and it was one of the first scientific descriptions of what had happened in killing centres such as Auschwitz. It also provided an analysis of the psychology of the SS-men who manned these camps and of their victims:the prisoners. At that time there was little interest in the Netherlands in recounting these events,but surprisingly the thesis was much in demand. It was later translated into English,Swedish and Japanese.

Willem Piso was a Dutch physician and naturalist who participated as an expedition doctor in Dutch Brazil from 1637 –1644,sponsored by count Johan Maurits van Nassau-Siegen and the Dutch West India Company. Piso became one of the founders of tropical medicine.
Barend Joseph Stokvis was a physician and professor of physiology and pharmacology at the University of Amsterdam. He is mainly remembered for his description of acute porphyria in 1889. As a researcher in chemical pathology he made contributions to the understanding of a number of diseases,such as diabetes. He was also considered an expert in tropical medicine and a celebrated medical educator. He authored an influential pharmacology textbook. Stokvis was one of a number of prominent 19th century Jewish physicians in the Netherlands.
The Nederlands Tijdschrift voor Geneeskunde is the main medical journal in the Netherlands,appearing weekly. Established in 1857,it is one of the world's oldest journals. Its publication language is exclusively Dutch. The journal is published and supported by the Vereniging NTvG,which is currently composed of 209 medical scientists. The current editor-in-chief is Yolanda van der Graaf. The Journal's headquarter is situated in Amsterdam,the Netherlands. From early on,the objective was to create an overarching and all-encompassing journal for medical professionals to exchange insights,knowledge and opinion,and to guarantee consistent progress throughout the country. At present,the main sections include:News,Opinion,Research,Clinical Practice,Perspective. Nowadays,the NTvG focuses on reviews and commentaries of research articles which are often published in English. Further,it continues to produce research of medical practice mainly in the Netherlands.

Hans Herman Cohen was a Dutch microbiologist. He was director-general of the Netherlands National Institute for Public Health and the Environment (RIVM) between 1984 and 1986. As a microbiologist Cohen worked on development of polio vaccines in the Netherlands.
Transient expression,more frequently referred to "transient gene expression",is the temporary expression of genes that are expressed for a short time after nucleic acid,most frequently plasmid DNA encoding an expression cassette,has been introduced into eukaryotic cells with a chemical delivery agent like calcium phosphate (CaPi) or polyethyleneimine (PEI). However,unlike "stable expression," the foreign DNA does not fuse with the host cell DNA,resulting in the inevitable loss of the vector after several cell replication cycles. The majority of transient gene expressions are done with cultivated animal cells. The technique is also used in plant cells;however,the transfer of nucleic acids into these cells requires different methods than those with animal cells. In both plants and animals,transient expression should result in a time-limited use of transferred nucleic acids,since any long-term expression would be called "stable expression."
Albert Zwaveling was a Dutch surgeon specializing in oncology,who worked as a professor at Leiden University between 1972 and 1992. He implemented chemotherapy in Dutch surgery and is considered to be one of the founders of surgical oncology in the Netherlands.
