This was the first edition of the tournament, primarily organised due to the cancellation of many tournaments during the 2020 season, because of the ongoing COVID-19 pandemic.

Ashleigh Barty was the defending champion from when the tournament was last held in 2019, and she successfully defended her title after Bianca Andreescu retired from the final with the scoreline at 6–3, 4–0. Barty saved a match point in the second round against Kristína Kučová en route to the title. The victory earned Barty her first successful title defense in her singles career and made her the sixth woman to accomplish a title defense at the Miami Open.

This was the first edition of the tournament.

Ons Jabeur defeated Jessica Pegula in the final, 7–5, 0–6, 6–2 to win the women's singles tennis title at the 2022 Madrid Open. It was her first WTA 1000 title, and she became the first Arab and African woman to win a WTA 1000 title. Pegula reached the final after saving a match point in her first round match against Camila Giorgi.
