County routes in Wayne County, New York, are only posted on street blade signs. Each route number is the product of two different numbering systems, both county-wide in nature. The last two digits of the route number serve as a road's base designation and are assigned sequentially from the Monroe County line in the west to the Cayuga County line in the east and from the Lake Ontario shoreline in the north to the Ontario and Seneca county lines in the south. The first digit of each route's designation indicates where the route is located: routes numbered in the 100s are north of Ridge Road; routes in the 200s are south of Ridge Road and north of New York State Route 31 (NY 31); and routes in the 300s are south of NY 31. If a road crosses either of those highways, the first digit of the route's number changes to reflect the road's location in the latter numbering grid.
Routes ending in an even number run from north to south; routes with an odd designation travel from east to west.

New York State Route 104 (NY 104) is a 182.41-mile-long (293.56 km) east–west state highway in Upstate New York in the United States. It spans six counties and enters the vicinity of four cities—Niagara Falls, Lockport, Rochester, and Oswego—as it follows a routing largely parallel to the southern shoreline of Lake Ontario, along a ridge of the old shoreline of Glacial Lake Iroquois. The western terminus of NY 104 is an intersection with NY 384 in Niagara Falls, Niagara County, while its eastern terminus is a junction with NY 13 in the town of Williamstown, Oswego County. The portion of NY 104 between Rochester and the village of Webster east of the city is a freeway known as the Keeler Street Expressway west of NY 590 and the Irondequoit–Wayne County Expressway east of NY 590; from Williamson to Oswego, NY 104 is a super two highway.
New York State Route 47 (NY 47) was a 19.34-mile (31.12 km) long state highway around Rochester in New York, in the United States. It formed a semicircle through the inner suburbs of Rochester, following expressways west and east of the city and surface streets south of Rochester. The western terminus of the route was at an interchange with NY 104 west of the city limits in Greece. The eastern terminus was at an intersection with Culver Road in Irondequoit close to Lake Ontario. In between its western and eastern extents, NY 47 met Interstate 490 (I-490) twice. The limited-access highway portions of the route were known as the Rochester Outer Loop in contrast to the Inner Loop around downtown Rochester.
New York State Route 18 (NY 18) is an east–west state highway in western New York in the United States. It runs parallel to the south shore of Lake Ontario for most of its length between Niagara County and Monroe County. NY 18, which also passes through Orleans County, acts as a northerly alternate to NY 104, another east–west route that parallels NY 18 to the south on Ridge Road. The western terminus of NY 18 is at a complex grade-separated interchange with NY 104 outside the village of Lewiston. Its eastern terminus is at a junction with NY 104 in an area of Rochester known as Eastman Business Park.
New York State Route 404 (NY 404) is an east–west state highway located in eastern Monroe County, New York, in the United States. It extends for just over 10 miles (16 km) from an interchange with NY 590 in Irondequoit to an intersection with NY 104 on the Monroe–Wayne County line in the town of Webster. The route traverses the southern tip of Irondequoit Bay and passes through the village of Webster, where NY 404 intersects NY 250. Most of NY 404 passes through commercial areas; however, the western and eastern extents of the highway serve areas more residential in nature. The westernmost 3 miles (5 km) of the route is part of the Seaway Trail, a National Scenic Byway.
New York State Route 279 (NY 279) is a north–south state highway in Orleans County, New York, in the United States. The southern terminus of the route is at an intersection with NY 98 north of the village of Albion in the town of Gaines. Its northern terminus is at a junction with NY 18 in Carlton near where NY 18 meets the western end of the Lake Ontario State Parkway. The entirety of NY 279 north of NY 104 is maintained by Orleans County; south of NY 104, the route is maintained by the New York State Department of Transportation. NY 279 is a rural connector highway that serves only one community, the hamlet of Waterport on the banks of Oak Orchard Creek in Carlton.
New York State Route 237 (NY 237) is a north–south state highway located in the western part of New York in the United States. The southern terminus of the route is at an intersection with NY 5 in Stafford. Its northern terminus is at an interchange with the Lake Ontario State Parkway immediately south of Lake Ontario in Kendall. NY 237 passes through mostly rural areas of Genesee and Orleans counties; however, it also passes through several small communities, including the village of Holley.
New York State Route 318 (NY 318) is an east–west state highway in the Finger Lakes region of New York in the United States. The western terminus of the route is at an intersection with NY 14 at New York State Thruway exit 42 in the town of Phelps. The eastern terminus is at an intersection with U.S. Route 20 (US 20) and NY 5 northeast of the hamlet of Seneca Falls. All but 0.70 miles (1.13 km) of the 10.90-mile (17.54 km) route is located in Seneca County.
New York State Route 31F (NY 31F) is an east–west state highway located in the vicinity of Rochester, New York, in the United States. The western terminus of the route is at an intersection with NY 96 north of the Monroe County village of Pittsford. Its eastern terminus is at a junction with NY 31 in the Wayne County hamlet of Macedon. Along the way, NY 31F serves the villages of East Rochester and Fairport and connects to Interstate 490 (I-490) in the town of Pittsford. From Fairport eastward, NY 31F roughly parallels the Erie Canal, which the route crosses over twice: once west of Fairport and again north of Macedon.
New York State Route 120A (NY 120A) is a state highway in southern Westchester County, New York, in the United States. It serves as an alternate route to the southern half of NY 120 running along the New York–Connecticut state line. The southern terminus of the route is at NY 120 in Harrison. Its northern terminus is at NY 120 in North Castle. Most of the 8.55-mile (13.76 km) route straddles the New York–Connecticut state line, and part of the route is physically located in Connecticut. However, this part of the highway is not recognized by Connecticut as "Route 120A", and is considered to be part of NY 120A by the New York State Department of Transportation (NYSDOT).
New York State Route 286 (NY 286) is an east–west state highway in the vicinity of the city of Rochester, New York, in the United States. It links Rochester to its eastern suburbs and to western Wayne County. The western terminus of the route is at exit 7 on NY 590 just inside the Rochester city limits. Its eastern terminus is at NY 350 in Walworth. NY 286 is named Browncroft Boulevard from NY 590 to its junction with Qualtrough and Clark roads in Penfield. This portion of the highway passes through predominantly residential areas. East of this intersection, the route is known as Atlantic Avenue and traverses more rural areas.
New York State Route 260 (NY 260) is a north–south state highway in Monroe County, New York, in the United States. The southern terminus of the route is at an intersection with NY 31 in the town of Sweden. Its northern terminus is at a junction with the Lake Ontario State Parkway in the town of Hamlin near the Lake Ontario shoreline. NY 260 crosses the Erie Canal near Brockport and intersects NY 104 and NY 18 in Clarkson. Much of NY 260 is a two-lane highway that passes through rural portions of western Monroe County.
New York State Route 350 (NY 350) is a north–south state highway in western Wayne County, New York, in the United States. It extends for 11.40 miles (18.35 km) from an intersection with NY 31 and NY 31F in the village of Macedon to a junction with NY 104 in the town of Ontario. NY 350 serves as the eastern terminus for three routes: the aforementioned NY 31F, NY 286, and NY 441, all of which originate in the Rochester area. Aside from the village of Macedon at its south end and the hamlet of Ontario Center at its north terminus, NY 350 serves mostly rural areas dominated by farmland. NY 350 was assigned c. 1932 to the portion of its routing south of Cator Corners, the north junction with NY 31F. It was extended northward to its current terminus in the 1940s.
New York State Route 295 (NY 295) is a 12.88-mile (20.73 km) state highway in Columbia County, New York, in the United States. It begins at an intersection with NY 66 in the village of Chatham and heads generally easterly to the Massachusetts border in the town of Canaan, where the road becomes Massachusetts Route 295 for another 1.67 miles (2.69 km) to a junction with Route 41 in Richmond, Massachusetts. Route 295 is the highest-numbered Massachusetts state highway, and the only state highway that shares its number with another numbered highway in Massachusetts, this being Interstate 295.
