Mendefera subregion is a subregion in the Debub (Southern) region (Zoba Debub) of Eritrea. Its capital lies at Mendefera.

The Southern region is an administrative region of Eritrea. The region was formed on 15 April 1996, from the historical provinces of Seraye and Akele Guzai. It lies along a portion of the national border with Ethiopia. As of 2005, the region had a population of 952,100 compared to a population of 839,700 in 2001. The net growth rate was 11.81 per cent. The total area of the province was 8000.00 km2 and the density was 119.01 persons per km2. Mendefera, formerly Adi Ugri, is an ancient town which is now the capital city of the Southern Region or Zoba Debub of Eritrea. The town's name derives from the high hill in the center of the city and it means that "who dared it" or "መን ደፈራ"( in Tigrinya) as it was a jungle and is a source of pride to Eritreans. The city is a reminder of the local anti-colonial movement.

Adi Quala is a market town in southern (Debub) part of Eritrea. It is located 32 km south of Mendefera nearly 25 km from the Ethiopian border, over 2,000m above sea level.

Metera or መጠራ (𐩣𐩷𐩧) is a small town and important archeological site located in the Debub Region of Eritrea. Situated a few kilometers south of Senafe (ሰንዓፈ), it was a major city in the Dʿmt (𐩵𐩲𐩣𐩩) and Aksumite kingdoms. The town has the oldest Example of Ge’ez, which is a pre-Aksumite Obelisk, Hawulti (monument). Since Eritrean independence, the National Museum of Eritrea has petitioned the Ethiopian government to return artifacts removed from the site. However, the efforts have thus far been rebuffed.

Adi-Ada/:ትግርኛ/ዓዲ ዓዳ is a village of the former historical province of Seraye, which is located 7 km southeast to the city of Mendefera in present Southern Region of the state of Eritrea. The village has an age of at least 500 years, which is composed of four families called Enda Tesfit, Enda Nayzgi, Enda Oqubay and Enda Andehans who are founding fathers of the village Adi-Ada which has more than 300 households and an estimated population of 1,500 people.

Adi-ugri or Adi-wegri is a village in the former province of Seraye, Eritrea. The village is located some two kilometers from the city center of Mendefera to the west and is the origin of Mendefera city.
