Buciclic acid, or bucyclic acid, systematic name trans-4-butylcyclohexane-1-carboxylic acid, is a simple alkyl-substituted cyclohexanecarboxylic acid. [1] The salts and esters of buciclic acid are known as buciclates (bucyclates). Pharmaceutical examples of esters of this acid include testosterone buciclate, a long-acting prodrug of the androgen testosterone, [2] and dimethandrolone buciclate, a prodrug of dimethandrolone.

Estradiol dienantate

Estradiol dienanthate (EDE), sold under the brand names Climacteron among others, is a long-acting estrogen medication which was previously used in menopausal hormone therapy for women and to suppress lactation in women. It was formulated in combination with estradiol benzoate (EB), a short-acting estrogen, and testosterone enanthate benzilic acid hydrazone (TEBH), a long-acting androgen/anabolic steroid. EDE has not been made available for medical use alone. The medication, in combination with EB and TEBH, was given by injection into muscle once or at regular intervals, for instance once every 6 weeks.

Testosterone valerate

Testosterone valerate, or testosterone pentanoate, also known as androst-4-en-17β-ol-3-one 17β-valerate, is a synthetic, steroidal androgen and an androgen ester – specifically, the C17β valerate ester of testosterone – which is used in veterinary medicine. It is administered via intramuscular injection and acts as a long-lasting prodrug of testosterone. The medication is available as a component of the veterinary drug Deposterona, which is marketed in Mexico and also contains testosterone acetate and testosterone undecanoate. Testosterone valerate is a short-to-medium duration ester of testosterone, with an elimination half-life of approximately twice that of the short-acting testosterone propionate.

Dimethandrolone buciclate

Dimethandrolone buciclate, or dimethandrolone bucyclate, also known as 7α,11β-dimethyl-19-nortestosterone 17β-buciclate, is a synthetic anabolic–androgenic steroid (AAS) and a derivative of nandrolone (19-nortestosterone) which was developed by the Contraceptive Development Branch (CDB) of the National Institute of Child Health and Human Development (NICHD) and has not been marketed at this time. It is an androgen ester – specifically, the C17β buciclate (4-butylcyclohexane-1-carboxylate) ester of dimethandrolone (7α,11β-dimethyl-19-nortestosterone) – and acts as a prodrug of dimethandrolone in the body. Dimethandrolone buciclate is or was under investigation as a potential male contraceptive.

11β-Methyl-19-nortestosterone dodecylcarbonate

11β-Methyl-19-nortestosterone 17β-dodecylcarbonate (11β-MNTDC) is a synthetic and orally active anabolic–androgenic steroid (AAS) and a derivative of nandrolone (19-nortestosterone) which was developed by the Contraceptive Development Branch (CDB) of the National Institute of Child Health and Human Development (NICHD) and has not been marketed for medical use at this time. It is an androgen ester – specifically, the C17β dodecylcarbonate ester of 11β-methyl-19-nortestosterone (11β-MNT) – and acts as a prodrug of 11β-MNT in the body.

Testosterone enantate benzilic acid hydrazone

Testosterone enantate benzilic acid hydrazone (TEBH), or testosterone 17β-enantate 3-benzilic acid hydrazone, is a synthetic, injected androgen/anabolic steroid and an androgen ester – specifically, the C17β enantate (heptanoate) ester and C3 benzilic acid hydrazone of testosterone. It was previously marketed in combination with estradiol benzoate and estradiol dienantate under the brand names Climacteron, Lactimex, and Lactostat. Clinical studies have assessed this formulation. TEBH was first described in the scientific literature in 1959. It is a very long-lasting prodrug of testosterone when administered in oil via intramuscular injection.

Polytestosterone phloretin phosphate

Polytestosterone phloretin phosphate (PTPP) is an androgen and anabolic steroid as well as androgen ester which was never marketed. It is an ester of testosterone with phosphoric acid that is in the form of a polymer and is coupled with phloretin. Like other androgen esters, PTPP acts as a long-lasting prodrug of testosterone in the body. However, analogously to the polymeric estrogen esters polyestradiol phosphate (PEP), polyestriol phosphate (PE3P), and polydiethylstilbestrol phosphate (PSP), PTPP has a strongly prolonged duration with very uniform testosterone levels in animals compared to non-polymeric testosterone esters. According to its developers, this is "exactly the effect which should be aimed at in order to approach natural hormone production as closely as possible". PTPP was developed around 1953 at the same time as PEP and its patent was published in 1960. The patent was assigned to the Swedish pharmaceutical company Leo Läkemedel AB, which also developed PEP.

EC586

EC586, also known as testosterone 17β-(1- -L-proline), is an androgen and anabolic steroid which is under development by Evestra for use in androgen replacement therapy in men. It is an orally active androgen ester – specifically, a C17β sulfonamide–proline ester of the natural and bioidentical androgen testosterone – and acts as a prodrug of testosterone in the body. However, unlike oral testosterone and conventional oral testosterone esters such as testosterone undecanoate, EC586 has high oral potency, may undergo little or no first-pass metabolism, and may not have disproportionate androgenic effects in the liver. As such, it may have a variety of desirable advantages over oral testosterone, similarly to parenteral testosterone, but with the convenience of oral administration. Evestra intends to seek Investigational New Drug status for EC586 in the fourth quarter of 2018.

Trestolone enanthate

Trestolone enanthate, also known as 7α-methyl-19-nortestosterone 17β-enanthate, is an androgen and anabolic steroid (AAS) and progestogen which was never marketed. It is an androgen ester; specifically, it is the C17β enanthate (heptanoate) ester of trestolone (7α-methylestr-4-en-17β-ol-3-one). Trestolone enanthate has low affinity for sex hormone-binding globulin (SHBG), similarly to testosterone enanthate.
