The 2012 Greater Western Sydney Giants season was the club's first season of senior competition in the Australian Football League (AFL). The club also fielded its reserves team in the North East Australian Football League (NEAFL) for its second season, but under the new name University of Western Sydney Giants.
Kevin Sheedy was the first head coach of the Giants, while Mark Williams was the assistant coach.

Mark McVeigh is a former professional Australian rules footballer who played for the Essendon Football Club in the Australian Football League (AFL). He served as the caretaker senior coach of the Greater Western Sydney Giants following the resignation of Leon Cameron in 2022.
Adam Kingsley is a former Australian rules footballer and current senior coach of the Greater Western Sydney Giants in the Australian Football League (AFL). Kingsley was a premiership player for Port Adelaide, before spending time as an assistant coach at St Kilda and Richmond.

He was one of three inaugural Greater Western Sydney co-captains, leading the club for eight seasons, and is the club's games record holder with 253 games.

He was listed with the Port Adelaide Football Club from 2006 to 2009, but did not play a senior game for the club. He returned to the AFL with expansion club Greater Western Sydney, making his debut in round 1 of the 2012 season. Giles moved on to Essendon for the 2015 season, and then spent two years with West Coast before retiring due to a degenerative knee condition. In total, he played 63 AFL games.

Curtly Hampton is a former professional Australian rules footballer who played for the Adelaide Football Club in the Australian Football League (AFL). He was originally recruited by incoming AFL club Greater Western Sydney with one of their Northern Territory zone selections in 2010. He was traded to Adelaide at the end of 2015, after 51 matches in four AFL seasons for Greater Western Sydney. Hampton is originally from Alice Springs, and is the son of former politician Karl Hampton.

Zachary Williams is a professional Australian rules footballer playing for the Carlton Football Club in the Australian Football League (AFL). A utility, 1.85 metres tall and weighing 84 kilograms, Williams also plays as a defender and mid-fielder. Williams previously represented the Greater Western Sydney Football Club, playing 113 games over eight seasons from 2013 to 2020. He was recruited by Greater Western Sydney as the 54th pick in the 2012 AFL Rookie Draft, making his debut in round five of the 2013 AFL season. Williams earned a nomination for the 2013 AFL Rising Star award and has played the 14th most games for Greater Western Sydney Club. At the end of the 2020 season, Williams signed with Carlton as a restricted free agent.
Jake Stringer is a professional Australian rules footballer playing for the Greater Western Sydney Giants in the Australian Football League (AFL). He previously played for the Western Bulldogs from 2013 to 2017 and for the Essendon Football Club from 2018 to 2024. Stringer was selected in the All-Australian team in 2015 and won an AFL premiership with the Bulldogs in 2016, the club's first in 62 years. He has also led his clubs' goalkicking on six occasions.
The Australian Football League registered the name Western Sydney Football Club Ltd with the Australian Securities and Investments Commission in January 2008. On 12 March 2008 the AFL received unanimous support from the existing 16 clubs for two expansion teams to enter the league, one of which was to be based in western Sydney. The Western Sydney working party met on 22 July to discuss player rules and draft concessions.

As of the finals series of the 2024 AFL season, the head-to-head score is in favour of the Sydney Swans with 19 wins to 10; the teams have also met four times in finals matches, with Greater Western Sydney winning three finals to Sydney's one.
Alicia Eva is an Australian rules footballer playing for the Greater Western Sydney Giants in the AFL Women's (AFLW). She previously played for the Collingwood Football Club in 2017. Eva was selected in the AFL Women's All-Australian team and won the Gabrielle Trainor Medal in her first season at the Giants in 2018. She served as Greater Western Sydney captain from 2020 to 2023, and is Greater Western Sydney's games record holder with 66 games.
The 2019 AFL Grand Final was an Australian rules football game contested between the Richmond Football Club and Greater Western Sydney Giants at the Melbourne Cricket Ground on 28 September 2019. It was the 124th annual grand final of the Australian Football League, staged to determine the premiers for the 2019 AFL season. The match, attended by 100,014 spectators, was won by Richmond by a margin of 89 points, marking the club's twelfth VFL/AFL premiership and their second in three seasons. Richmond's Dustin Martin won the Norm Smith Medal as the player judged best on ground.
