Barbara Imperiali (b. January 1, 1957) is a Professor of Biology and Chemistry at Massachusetts Institute of Technology and Affiliate Member of the Broad Institute. She is an elected member of the National Academy of Sciences and a Fellow of the Royal Society of Chemistry.

Education and early career

Barbara Imperiali was raised in England and attended the Southbank School in Caterham in Surrey, followed by Coloma Grammar School in Croydon where she specialized in studying the sciences. [1] She received her Bachelor of Science in Medicinal Chemistry from the University College London in 1979. She attended Massachusetts Institute of Technology, where she received her Doctor of Philosophy in synthetic organic chemistry in 1983. She joined the lab of Satoru Masamune, working to develop and apply new techniques to synthesize a class of antibiotic chemicals known as ansamycin. [2] As a postdoc in the Masamune lab she worked with the enzyme beta-ketothiolase. [1]

She did additional postdoctoral research at Brandeis University in the laboratory of Robert Abeles, where she received training in chemical biology designing, synthesizing, and evaluating peptide-based protease inhibitors. [1]

Research

In 1986, Imperiali became an Assistant Professor at Carnegie Mellon University before moving her laboratory to California Institute of Technology in 1989. While at Caltech, she was influenced by the research of Dennis Dougherty, who was applying unnatural amino acid mutagenesis to understand interactions of ligand-gated membrane channels and receptors. Imperiali's work became focused on problems at the chemistry–biology interface, using chemistry to better understand the structure and function of proteins in vivo. [1]

In 1999, she returned to MIT as the Ellen Swallow Richards Professor of Biology. [1] Her research program is focused on protein glycosylation, a process by which a carbohydrate is added to a protein to change its structure or function. Her lab has developed techniques to track and understand how glycosylation at the time of protein production—a process known as translation—affects protein folding and conformation. Applying techniques such as Fluorescence Resonance Energy Transfer and nuclear magnetic resonance, she has elucidated how glycosylation might protect proteins against misfolding and how it affects folding mechanics and thermodynamic stability. In addition to studying glycosylation, Imperiali has also led investigations into how to design, develop, and apply chemical approaches to understand and manipulate biochemical processes for a variety of applications. Her group has collaborated to develop a fluorescence-based method to detect phosphorylation of peptides and proteins to monitor the activity of kinases, which are enzymes that catalyze phosphorylation and coordinate a vast array of cellular activities. [5] This method can be applied to screen for protein kinase inhibitors and monitor their activities in cells and tissues, enabling tracking of signaling pathways and providing a potential diagnostic tool for diseases like cancers that may result from disrupted kinase-mediated regulation. [6]

She has also authored the textbook Chemical Glycobiology. [7]

<i>O</i>-GlcNAc

O-GlcNAc is a reversible enzymatic post-translational modification that is found on serine and threonine residues of nucleocytoplasmic proteins. The modification is characterized by a β-glycosidic bond between the hydroxyl group of serine or threonine side chains and N-acetylglucosamine (GlcNAc). O-GlcNAc differs from other forms of protein glycosylation: (i) O-GlcNAc is not elongated or modified to form more complex glycan structures, (ii) O-GlcNAc is almost exclusively found on nuclear and cytoplasmic proteins rather than membrane proteins and secretory proteins, and (iii) O-GlcNAc is a highly dynamic modification that turns over more rapidly than the proteins which it modifies. O-GlcNAc is conserved across metazoans.

Marcos Boris Rotman was a Chilean American immunologist–molecular biologist and professor emeritus of Medical Science at Alpert Medical School of Brown University. He is widely recognized for performing the first single molecule experiments in biology. He died in July 2021 at the age of 96.

Sarah E. O'Connor is an American molecular biologist working to understand the molecular machinery involved in assembling important plant natural products – vinblastine, morphine, iridoids, secologanin – and how changing the enzymes involved in this pathway lead to diverse analogs. She was a Project Leader at the John Innes Centre in the UK between 2011 and 2019. O'Connor was appointed by the Max Planck Society in 2018 to head the Department of Natural Product Biosynthesis at the Max Planck Institute for Chemical Ecology in Jena, Germany, taking up her role during 2019.

Mei Hong is a Chinese-American biophysical chemist and professor of chemistry at the Massachusetts Institute of Technology. She is known for her creative development and application of solid-state nuclear magnetic resonance (ssNMR) spectroscopy to elucidate the structures and mechanisms of membrane proteins, plant cell walls, and amyloid proteins. She has received a number of recognitions for her work, including the American Chemical Society Nakanishi Prize in 2021, Günther Laukien Prize in 2014, the Protein Society Young Investigator award in 2012, and the American Chemical Society’s Pure Chemistry award in 2003.

Charles Clifton Richardson is an American biochemist and professor at Harvard University. Richardson received his undergraduate education at Duke University, where he majored in medicine. He received his M.D. at Duke Medical School in 1960. Richardson works as a professor at Harvard Medical School, and he served as editor/associate editor of the Annual Review of Biochemistry from 1972 to 2003. Richardson received the American Chemical Society Award in Biological Chemistry in 1968, as well as numerous other accolades.

James Allen Wells is a Professor of Pharmaceutical Chemistry and Cellular & Molecular Pharmacology at the University of California, San Francisco (UCSF) and a member of the National Academy of Sciences. He received his B.A. degrees in biochemistry and psychology from University of California, Berkeley in 1973 and a PhD in biochemistry from Washington State University with Ralph Yount, PhD in 1979. He completed his postdoctoral studies at Stanford University School of Medicine with George Stark in 1982. He is a pioneer in protein engineering, phage display, fragment-based lead discovery, cellular apoptosis, and the cell surface proteome.

Julie Anne Champion is a professor of chemistry who holds the William R. McLain Endowed Term Professorship at Georgia Institute of Technology. She is known for her work on biomaterials used for drug delivery.
