Sir Hugh Reginald Brentnall Pelham, FRS FMedSci [6] [7] (born 26 August 1954) [1] is a cell biologist who has contributed to our understanding of the body's response to rises in temperature through the synthesis of heat shock proteins. [7] He served as director of the Medical Research Council (MRC) Laboratory of Molecular Biology (LMB) between 2006 and 2018. [8]
Pelham was educated at Marlborough College in Marlborough, Wiltshire and Christ's College, Cambridge. He graduated with a Master of Arts degree in Natural Sciences followed by a PhD for research on transcription and translation in immature blood cells (Reticulocytes). [9] His PhD was supervised by Richard J. Jackson and Tim Hunt, [7] who went on to receive the Nobel Prize in Physiology or Medicine in 2001.
Pelham is an authority on the movement of proteins within cells. Pelhams's work has explained how some proteins can protect cells from damage. He has also shown how cells remove damaged or unwanted proteins – vital for maintaining their healthy functioning. More recently, his research investigates how proteins are modified and sorted to their correct places within cells and aims to find ways of blocking these processes. [7] [10] [11] [12] [13]
Pelham has been a visiting professor at the University of Zurich and held many posts at the Laboratory of Molecular Biology in Cambridge, where he succeeded Richard Henderson to become the LMB's Director in 2006. [7] [8] He has been an Honorary Professor of Molecular Biology at the University of Cambridge since 2015. [1]
Pelham was knighted by Elizabeth II in the 2011 Birthday Honours and elected a Fellow of the Royal Society (FRS) in 1988. [7] His certificate of election reads:
Distinguished for his contributions to protein biosynthesis, the control of gene activity and intracellular sorting. He developed a sensitive in vitro translation system, with which he discovered that naturally "leaky" termination codons exist in plant virus RNAs, and achieved the first correct synthesis and processing of viral polyproteins in vitro. He showed that the transcription factor TFIIIA, which is required in Xenopus oocytes for 5S rDNA transcription, binds to the gene product, %S RNA and is present in large amounts in oocytes. From studies on heat shock genes, he identified the first regulatory DNA sequence (the "Pelham" box) in a eukaryotic gene, proving this alone could confer heat inducibility on another gene. He has shown that this sequence is the binding site for a transcription factor which is modified by heat shock, thus establishing the basic mechanism of induction of these genes. He has clarified the function of heat shock proteins, finding that two of these reside in the lumen of the endoplasmic reticulum. This led to his discovery that a C-terminal amino acid sequence is a novel sorting signal, preventing proteins from being exported from the lumen of the endoplasmic reticulum. [14]
Pelham gave the Florey Lecture in 1992, was elected a Fellow of the Academy of Medical Sciences (FMedSci) in 1998. [6] In 1999 he gave the Croonian Lecture and he was awarded the King Faisal International Prize in 1996. [3] He won the Louis-Jeantet Prize for Medicine in 1991 and the EMBO Gold Medal in 1989. He was awarded the Colworth Medal from the Biochemical Society in 1988 and elected a member of the European Molecular Biology Organization (EMBO) in 1985. [15]
Robert G. Roeder is an American biochemist. He is known as a pioneer scientist in eukaryotic transcription. He discovered three distinct nuclear RNA polymerases in 1969 and characterized many proteins involved in the regulation of transcription, including basic transcription factors and the first mammalian gene-specific activator over five decades of research. He is the recipient of the Gairdner Foundation International Award in 2000, the Albert Lasker Award for Basic Medical Research in 2003, and the Kyoto Prize in 2021. He currently serves as Arnold and Mabel Beckman Professor and Head of the Laboratory of Biochemical and Molecular Biology at The Rockefeller University.
Sir Richard Timothy Hunt, is a British biochemist and molecular physiologist. He was awarded the 2001 Nobel Prize in Physiology or Medicine with Paul Nurse and Leland H. Hartwell for their discoveries of protein molecules that control the division of cells. While studying fertilized sea urchin eggs in the early 1980s, Hunt discovered cyclin, a protein that cyclically aggregates and is depleted during cell division cycles.
Kim Ashley Nasmyth is an English geneticist, the Whitley Professor of Biochemistry at the University of Oxford, a Fellow of Trinity College, Oxford, former scientific director of the Research Institute of Molecular Pathology (IMP), and former head of the Department of Biochemistry, University of Oxford. He is best known for his work on the segregation of chromosomes during cell division.
In molecular biology, heat shock factors (HSF), are the transcription factors that regulate the expression of the heat shock proteins. A typical example is the heat shock factor of Drosophila melanogaster.
Mariann Bienz, Lady Pelham FRS FMedSci is a Swiss-British molecular biologist based at the UK Medical Research Council Laboratory of Molecular Biology. She has been a member of their Senior Scientific Staff since 1991, was Joint-head of Cell Biology in 2007-08 and has been a Group Leader of Protein and Nucleic Acid Chemistry Division since 2008.
Christopher John Marshall FRS FMedSci was a British scientist who worked as director of the Division for Cancer Biology at the Institute of Cancer Research. Marshall was distinguished for research in the field of tumour cell signalling. His track record includes the discovery of the N-Ras oncogene , the identification of farnesylation of Ras proteins, and the discovery that Ras signals through the MAPK/ERK pathway. These findings have led to therapeutic development of inhibitors of Ras farnesylation, MEK and B-Raf.
Sir Adrian Peter Bird, is a British geneticist and Buchanan Professor of Genetics at the University of Edinburgh. Bird has spent much of his academic career in Edinburgh, from receiving his PhD in 1970 to working at the MRC Mammalian Genome Unit and later serving as director of the Wellcome Trust Centre for Cell Biology. His research focuses on understanding DNA methylation and CpG islands, and their role in diseases such as Rett syndrome.
George Gow Brownlee FRS FMedSci is a British pathologist and Fellow of Lincoln College, Oxford.
Dame Amanda Gay Fisher is a British cell biologist and Director of the Medical Research Council (MRC) London Institute of Medical Sciences at the Hammersmith Hospital campus of Imperial College London, where she is also a Professor leading the Institute of Clinical Sciences. She has made contributions to multiple areas of cell biology, including determining the function of several genes in HIV and describing the importance of a gene's location within the cell nucleus.
Ketan Jayakrishna Patel is a British-Kenyan scientist who is Director of the MRC Weatherall Institute of Molecular Medicine and the MRC Molecular Haematology Unit at the University of Oxford. His major discoveries have concerned the mechanisms and physiological importance of protein degradation in cells.
