Defending champion Roger Federer defeated Andre Agassi in the final, 6–3, 2–6, 7–6(7–1), 6–1 to win the men's singles tennis title at the 2005 US Open. It was his second US Open title and sixth major title overall. This was Agassi's last appearance in a major final.

This was the first US Open in which future champions Novak Djokovic, Andy Murray and Stan Wawrinka competed in the main draw. Noteworthy in this tournament was a strong performance by American men, despite 2003 champion and former world No. 1 Andy Roddick losing in the first round. Wildcard James Blake reached the quarterfinals, upsetting world No. 2 Rafael Nadal in the third round before losing to Agassi. Robby Ginepri, whose run was enhanced by Roddick's early defeat, reached the semifinals and also lost to Agassi.

This was also the last major tournament for former world No. 6 and French Open champion Albert Costa.

Marat Safin defeated Lleyton Hewitt in the final, 1–6, 6–3, 6–4, 6–4 to win the men's singles tennis title at the 2005 Australian Open. It was his second and last major title, having also won the 2000 US Open. Safin saved a match point en route to the title, against Roger Federer in the semifinals. Hewitt was the first Australian to reach the final since Pat Cash in 1988.

Rafael Nadal defeated Mariano Puerta in the final, 6–7(6–8), 6–3, 6–1, 7–5 to win the men's singles tennis title at the 2005 French Open. It was his first major title, the first of a record 14 French Open titles, and the first of a record 22 major men's singles titles overall. It was his second French Open title and second major title overall. It was the first of three consecutive years Nadal and Federer would contest the French Open final. It was the longest men's singles major final in history with 77 games, breaking the record of 71 games set at the 1927 Australian Open. The match also had the longest fifth set (16–14) in a major men's singles final, surpassing the 11–9 fifth set in the 1927 French Open final. The match took 4 hours and 17 minutes to complete, and the fifth set alone lasted 95 minutes. It was a repeat of the 2004 and 2005 finals, where Federer also beat Roddick to win the title. This was Federer's 11th grass-court title, surpassing the previous record held by Sampras (10). Notably, Roddick lost despite only having his serve broken once, in the last game of the match. By winning the title, Federer regained the world No. 1 singles ranking.

Juan Martín del Potro defeated the five-time defending champion Roger Federer in the final, 3–6, 7–6(7–5), 4–6, 7–6(7–4), 6–2 to win the men's singles tennis title at the 2009 US Open. He became the first Argentine man to win the title since Guillermo Vilas in 1977, and the first to win a major since Gastón Gaudio in the 2004 French Open. This was del Potro's only major title, although he would reach a second final in 2018. This was also the first US Open final since 1999 to go to five sets.

Roger Federer defeated Andy Murray in the final, 6–3, 6–4, 7–6(13–11) to win the men's singles tennis title at the 2010 Australian Open. It was his fourth Australian Open title and 16th major title overall.

Novak Djokovic defeated the defending champion Rafael Nadal in a rematch of the previous year's final, 6–2, 6–4, 6–7(3–7), 6–1 to win the men's singles tennis title at the 2011 US Open. It was his first US Open title, his third major of the year, and his fourth major title overall. Djokovic saved match points en route to the title, saving two against Roger Federer in the semifinals. This was the second consecutive US Open where Djokovic saved two match points against Federer to reach the final, and the fifth consecutive US Open where Djokovic and Federer played each other. With the loss, Federer failed to win a major in a calendar year for the first time since 2002.

Andy Murray defeated the defending champion Novak Djokovic in the final, 7–6(12–10), 7–5, 2–6, 3–6, 6–2 to win the men's singles tennis title at the 2012 US Open. It was his first major title, and he became the first British man to win a major since Fred Perry in 1936, having been runner-up on four previous occasions. The final lasted 4 hours and 54 minutes, an Open Era record-equaling US Open final duration.
