Italian writer and journalist Italo Calvino was the Jury President of the main competition. The Golden Lion winner was Marianne and Juliane directed by Margarethe von Trotta.

Emir Kusturica is a Bosnian-born Serbian film director, screenwriter, actor, producer and musician. Kusturica is one of the most distinguished European filmmakers active since the mid-1980s, best known for surreal and naturalistic movies that express deep sympathies for people from the margins.
Margarethe von Trotta is a German film director, screenwriter, and actress. She has been referred to as a "leading force" of the New German Cinema movement. Von Trotta's extensive body of work has won awards internationally. She was married to and collaborated with director Volker Schlöndorff. Although they made a successful team, von Trotta felt she was seen as secondary to Schlöndorff. Subsequently, she established a solo career for herself and became "Germany's foremost female film director, who has offered the most sustained and successful female variant of Autorenkino in postwar German film history". Certain aspects of von Trotta's work have been compared to Ingmar Bergman's features from the 1960s and 1970s.
The 62nd annual Venice International Film Festival opened on 31 August 2005 with Tsui Hark's Seven Swords and closed on 10 September 2005 with a screening of Peter Ho-sun Chan's musical Perhaps Love. The lineups were announced by the festival director Marco Müller on 28 July 2005 in Rome. The digital films competed in all categories for the first time of the festival history.

Marianne and Juliane, also called The German Sisters in the United Kingdom, is a 1981 West German film directed by Margarethe von Trotta. The screenplay is a fictionalized account of the true lives of Christiane and Gudrun Ensslin. Gudrun, a member of The Red Army Faction, was found dead in her prison cell in Stammheim in 1977. In the film, von Trotta depicts the two sisters Juliane (Christine) and Marianne (Gudrun) through their friendship and journey to understanding each other. Marianne and Juliane was von Trotta's third film and solidified her position as a director of the New German Cinema.

American actor and film director Al Pacino was presented with the Glory to the Film-maker award on 4 September, prior to the premiere of his upcoming film Wilde Salomé. Marco Bellocchio was awarded with the Golden Lion for Lifetime Achievement in September. The festival opened with the American film The Ides of March, directed by George Clooney, and closed with Damsels in Distress by Whit Stillman.

Leon Hirszman was a Brazilian film director, producer and screenwriter, and one of the main figures of Cinema Novo. He is best known for directing the 1981 film They Don't Wear Black Tie which won the Special Jury Prize at the 38th Venice International Film Festival. His other films include Fernanda Montenegro's 1965 film debut The Deceased, the 1972 adaptation of Graciliano Ramos's S. Bernardo and the documentary ABC da Greve. He died from complications of HIV/AIDS at 49.
