The following is a list of teams and cyclists that took part in the 2021 Giro d'Italia. [1]
Twenty-three teams received invitations to participate in the 2021 Giro d'Italia. All nineteen UCI WorldTeams are entitled and obliged to enter the race, and they will be joined by four second-tier UCI ProTeams. Alpecin–Fenix, the best performing UCI ProTeam in 2020, received an automatic invitation, while the other three teams were selected by RCS Sport, the organizers of the Giro. Bardiani–CSF–Faizanè and Vini Zabù continue their streak of wild card invitations to the race, while Eolo–Kometa will make its Grand Tour debut. [2] The teams were announced on 10 February 2021. On 15 April 2021, after positive doping tests for Matteo Spreafico and Matteo De Bonis left them facing a potential suspension, Vini Zabù withdrew their participation from the race. [5] In place of them, regular invitee Androni Giocattoli–Sidermec, which had initially been snubbed by race organizers, were awarded the last wildcard invitation. [6]

Vini Zabù was an Italian UCI ProTeam that existed from 2009 until 2021.
Manuel Belletti is an Italian former professional road bicycle racer, who last rode for UCI ProTeam Eolo–Kometa. He took a total of 21 professional wins during his career, nine of them on Italian soil.
Marco Frapporti is an Italian racing cyclist, who last rode for UCI ProTeam Vini Zabù.
Luca Wackermann is an Italian cyclist, who last rode for UCI ProTeam Team Polti Kometa. In October 2020, he was named in the startlist for the 2020 Giro d'Italia. However, at the end of stage 4, Wackermann was involved in a crash caused by a low-flying helicopter that blew some crash barriers into him. Wackermann suffered a broken nose and a suspected broken back in the incident.
Mattia Frapporti is an Italian former racing cyclist, who competed as a professional from 2014 to 2021. He rode at the 2014 UCI Road World Championships.
Matteo Malucelli is an Italian cyclist, who currently rides for UCI Continental team Bingoal WB.
Luca Pacioni is an Italian cyclist, who currently rides for UCI ProTeam Team Polti Kometa.
Matteo Spreafico is an Italian cyclist, who is provisionally suspended from the sport, following a positive drugs test.
Team Polti Kometa is an Italian-registered, Spanish-based UCI ProTeam cycling team, that was founded in 2018. The team was promoted from the UCI Continental level in 2021.
Andrea Garosio is an Italian cyclist, who currently rides for UCI ProTeam Team Polti Kometa. In May 2019, he was named in the startlist for the 2019 Giro d'Italia.
Simone Bevilacqua is an Italian former cyclist, who competed as a professional from 2018 to 2023. In October 2020, he was named in the startlist for the 2020 Giro d'Italia.
The 2021 Giro d'Italia was the 104th edition of the Giro, a three-week Grand Tour cycling stage race. It started on 8 May and finished on 30 May. On 26 May 2019, the race organisers RCS Sport originally announced that the start of the 2021 Giro would be in Sicily, Italy. However, on 4 February 2021, RCS Sport announced that the race would instead start in Turin, with the rest of the race route being announced on 24 February 2021.

The 2021 Giro di Sicilia was the 25th edition of the Giro di Sicilia road cycling stage race and the second edition since its revival in 2019. The race was originally scheduled for April but had been postponed due to COVID-19 pandemic restrictions, which had also caused the cancellation of the 2020 edition.
The 2021 Giro dell'Emilia was the 104th edition of the Giro dell'Emilia road cycling one day race in the titular region of central Italy. It was held on 2 October 2021 as part of the 2021 UCI Europe Tour and the 2021 UCI ProSeries calendars.
The 2021 Tre Valli Varesine was the 100th edition of the Tre Valli Varesine road cycling one day race, which was held in the Lombardy region of northwestern Italy on 5 October 2021. This edition was the race's first in the UCI ProSeries; the 2020 edition was expected to feature in the inaugural UCI ProSeries but was cancelled due to the COVID-19 pandemic. The race was the second of the 2021 Trittico Lombardo, which also included the Coppa Bernocchi held the day before and the Coppa Ugo Agostoni, to be held six days later.
