The first heats of the Women's 100 m Butterfly took place on the morning of Sunday, 26 July. The semi-final took place in the evening session of 26 July and the final took place on the evening of 27 July at the Foro Italico in Rome, Italy. In the final, 7 of the 8 swimmers finished within the same second, and 5 within the same 0.1 of a second.

Daiya Seto is a Japanese professional swimmer who specializes in individual medley, butterfly, breaststroke, and freestyle events. He holds the world record in the short course 400-metre individual medley and formerly held the world record in the short course 200-metre butterfly. He won the gold medal in the 400-metre individual medley at the 2012, 2014, 2016, 2018, 2021, and 2022 world short course championships, as well as at the 2013, 2015, and 2019 world long course championships.

Evgeny Mikhailovich Rylov is a Russian competitive swimmer and Olympic champion specializing in backstroke events. He won three gold medals at the 2014 Summer Youth Olympics in Nanjing, and a bronze medal at his senior international debut at the 2015 World Championships in Kazan. He also won a bronze medal at the 2016 Summer Olympics in Rio de Janeiro and a gold medal at the 2017 World Championships in Budapest, both were in the 200 metre backstroke event. In 2018, at the 2018 World Short Course Championships, he won gold medals in the 200 metre backstroke and 50 metre backstroke. At the 2019 World Championships, he won a gold medal in the 200 metre backstroke, silver medal in the 100 metre backstroke, and silver medal in the 50 metre backstroke. Michael Charles Andrew is an American competitive swimmer and an Olympic gold medalist. He was the 2016 world champion in the 100 meter individual medley. At his first Olympic Games, the 2020 Summer Olympics, he won a gold medal and set a world record as part of the 4x100 meter medley relay, placed fourth in the 100 meter breaststroke, fourth in the 50 meter freestyle, and fifth in the 200 meter individual medley. Andrew's swims in 2021 at the 2020 Olympics made him the first swimmer to represent the United States at an Olympic Games in an individual breaststroke event as well as another individual event other than an individual medley in the then-125-year-history of swimming at the Summer Olympics. Lillia Camille King is an American swimmer who specializes in breaststroke. She currently represents the Cali Condors, a team that is part of the International Swimming League. At the 2016 Summer Olympics, she won the gold medal in the 100-meter breaststroke competition and also won a gold medal in the 4x100 meter medley relay, in which she swam the breaststroke leg. At the 2020 Summer Olympics, King won a silver medal in the 4x100 meter medley relay for her efforts in the prelims, the silver medal in the 200-meter breaststroke, and the bronze medal in the 100-meter breaststroke. She is the current world record holder in the long course 100-meter breaststroke.

Blake John Pieroni is a three-time Olympic gold medalist in swimming. He is a two time Olympian and gold medalist in the 4×100-meter freestyle relay at both the 2016 Summer Olympics and the 2020 Summer Olympics. On the relay in 2016, he swam in the prelims of the race, on the 2020 Olympics 4×100-meter freestyle relay he swam in both the prelims and the final. Dylan Carter is a Trinidad and Tobago competitive swimmer. At the 2021 World Short Course Championships, he became the first swimmer representing Trinidad and Tobago to win a silver medal a World Short Course Championships, winning the silver medal in the 50 metre butterfly. Kristóf Milák is a Hungarian swimmer. He is the current holder of the world record in the long course 200 metre butterfly and the European record in the long course 100 metre butterfly. He also currently holds the Hungarian record in the short course 50 metre backstroke. At his first Olympic Games, the 2020 Summer Olympics, he won a gold medal in the 200 metre butterfly and a silver medal in the 100 metre butterfly. He has also won three gold medals and one silver at the World Aquatic Championships, as well as three gold medals at the European Aquatics Championships. Zachary "Zach" Douglas Apple is a retired American competitive swimmer who specialized in the sprint freestyle events. He used to swim for DC Trident in the International Swimming League. He won his first Olympic gold medal in the 4x100-meter freestyle relay at the 2020 Summer Olympics, swimming in the prelims and the final of the event, and later in the same Olympic Games won a gold medal and helped set a new world record and Olympic record in the 4x100-meter medley relay, swimming the freestyle leg of the relay in the final.

Rebecca Meder is a South African swimmer. She is the African record holder in the 100 metre individual medley and the South African record holder in the long course and short course 200 metre individual medley. At the 2022 World Short Course Championships, she placed sixth in the final of the 100 metre individual medley. In the 200 metre individual medley at the 2022 Commonwealth Games, she placed fourth in the final. At the 2021 African Championships, she won 11 gold medals, six in individual events and five in relay events, swimming on the finals relay for each of the five relay events.

David Popovici is a Romanian competitive swimmer who specializes in freestyle events. He is a former world record holder in the long course 100-metre freestyle and the current junior world record holder in the long course 200-metre freestyle. He also holds the junior world record in the short course 100-metre freestyle. At the 2022 World Aquatics Championships, Popovici won gold medals in his two signature events, becoming the first swimmer to do so at the same edition of the World Championships, since Jim Montgomery in 1973. Following his performances during the 2022 long course season, in which he won eleven gold medals and two silver medals, Popovici was voted Male Swimmer of the Year by both SwimSwam and Swimming World Magazine. He is the Americas record holder in the long course 400 meter freestyle and the American record holder in the 500 yard freestyle. In the 400 meter freestyle, he won the bronze medal at the 2020 Summer Olympics and the gold medal at the 2022 World Short Course Championships. Andrew Patrick “Drew” Kibler is an American competition swimmer specializing in freestyle events. He is a world record holder in the short course 4×200 meter freestyle relay and a former American record holder in the 4×200 yard freestyle relay. He won a gold medal in the 4×200 meter freestyle relay each at the 2022 World Aquatics Championships and the 2022 World Short Course Championships and placed fourth in the event at the 2020 Summer Olympics. In the 200 meter freestyle, he took fourth-place at both the 2022 World Aquatics Championships and the 2022 World Short Course Championships. He is also a three-time NCAA champion in the 4×200 yard freestyle relay, winning NCAA titles in the event in 2019, 2021, and 2022.

At the 2020 Summer Olympics, he won a gold medal in the 4×100 meter medley relay, swimming backstroke in the preliminaries, and placed ninth in the 100 meter backstroke.
