Jannik Sinner is the defending champion. ^[1]

Sinner and Carlos Alcaraz are in contention for the world No. 1 singles ranking. Sinner must achieve a better result than Alcaraz to retain the top spot. ^[2]

Novak Djokovic is attempting to win a record 25th Grand Slam singles title. ^[3] With his third-round win, Djokovic recorded his 192nd career match win at hardcourt majors, surpassing Roger Federer's record. ^[4] Djokovic was the oldest man to reach the quarterfinals of the US Open since Jimmy Connors in 1991, the oldest man to reach the quarterfinals of any major since Federer at the 2021 Wimbledon Championships, and the oldest man in the Open Era to reach all four major quarterfinals in a season. ^[5]

This marked the first time since 2010 that Grigor Dimitrov did not participate at a major, ending his streak of 58 consecutive major appearances. ^[6]

Coleman Wong became the first man from Hong Kong to qualify for a major main draw in the Open Era, ^{[7] [8]} and the first to win a match at a major singles main draw since Ip Koon Hung and Edwin Tsai at the 1954 Wimbledon Championships—when Hong Kong was a British colony. ^[9] Wong was also one of five qualifiers (with Leandro Riedi, Jan-Lennard Struff, Ugo Blanchet, and Jérôme Kym) to reach the third round, ^[10] tied for the highest number in a single edition at the US Open in men's singles in the Open Era with 1984 and 2021. ^[11]

Ranked No. 435 (and using protected ranking to enter the qualifying draw), Riedi was the lowest-ranked player to reach the fourth round of a major since Richard Krajicek at the 2002 Wimbledon Championships, and the lowest-ranked player to do so at the US Open since Jay Berger in 1985. ^[12]
