In enzymology, a phosphoglycerol geranylgeranyltransferase (EC 2.5.1.41) is an enzyme that catalyzes the chemical reaction

geranylgeranyl diphosphate + sn-glyceryl 1-phosphate diphosphate + sn-3-O-(geranylgeranyl)glyceryl 1-phosphate

Thus, the two substrates of this enzyme are geranylgeranyl diphosphate and sn-glyceryl 1-phosphate, whereas its two products are diphosphate and sn-3-O-(geranylgeranyl)glyceryl 1-phosphate.

This enzyme belongs to the family of transferases, specifically those transferring aryl or alkyl groups other than methyl groups. The systematic name of this enzyme class is geranylgeranyl diphosphate:sn-glyceryl phosphate geranylgeranyltransferase. Other names in common use include glycerol phosphate geranylgeranyltransferase, geranylgeranyl-transferase, and prenyltransferase.

Prenylation Addition of hydrophobic moieties to proteins or other biomolecules

Prenylation is the addition of hydrophobic molecules to a protein or a biomolecule. It is usually assumed that prenyl groups (3-methylbut-2-en-1-yl) facilitate attachment to cell membranes, similar to lipid anchors like the GPI anchor, though direct evidence of this has not been observed. Prenyl groups have been shown to be important for protein–protein binding through specialized prenyl-binding domains.

Geranylgeranyltransferase type 1 or simply geranylgeranyltransferase is one of the three enzymes in the prenyltransferase group. In specific terms, Geranylgeranyltransferase adds a 20-carbon isoprenoid called a geranylgeranyl group to proteins bearing a CaaX motif: a four-amino acid sequence at the carboxyl terminal of a protein. Geranylgeranyltransferase inhibitors are being investigated as anti-cancer agents.
