Maryland Route 316 is a state highway in the U.S. state of Maryland. Known as Appleton Road, the highway runs 2.67 miles (4.30 km) from MD 279 near Elkton north to MD 277 near Elk Mills in northeastern Cecil County. MD 316 was constructed in the early 1910s from Elkton to Elk Mills and in the early 1920s north of Elk Mills. In the early 1960s, the disjoint northern segment of the highway was transferred to the county and the highway's present southern terminus was established when MD 279 moved to a new alignment north of Elkton in the early 1960s.

Highway 316 is a highway in the Canadian province of Saskatchewan. It runs from Highway 16 near Clavet to Highway 5. Highway 316 is about 17 km (11 mi.) long. This area is an industrial commercial area along Highway #316 which extends from Clavet north to the PCS Patience Lake Mine which is just 10 miles from the city of Saskatoon. Highway 316 is a primary weight highway, and also runs near the Canadian National Railway line which goes from Saskatoon through Clavet. In 1941, the Canadian Pacific Railway ran from Saskatoon through Cheviot and on south eastward.

Pennsylvania Route 316 is a 16.6-mile-long (26.7 km) state highway located in Franklin County, Pennsylvania. The southern terminus is at the Maryland state line near Waynesboro, where the road continues into that state as Maryland Route 60. The northern terminus is at U.S. Route 11 in Chambersburg. PA 316 heads north from the state line through rural land to Waynesboro, where it forms a concurrency with PA 16. From here, the route continues through farmland, intersecting PA 914 south of New Franklin. PA 316 reaches Chambersburg and runs through developed area, coming to an interchange with Interstate 81 (I-81) before ending at US 11.
