It took place at the Estadio Mario Alberto Kempes in Córdoba, Argentina, from 20 February until 28 February 2021.

Matteo Berrettini defeated Aslan Karatsev in the final, 6–1, 3–6, 7–6(7–0), to win the men's singles tennis title at the 2021 Serbia Open.

Miomir Kecmanović was the defending champion, but chose to compete at the Summer Olympics instead.
