Due to the 2022 FIFA World Cup, the regular season began early on February 26, 2022, and ended on October 9. The team ended their season on October 15, 2022, after losing to FC Cincinnati 1–2 in the MLS Cup Playoffs.

The 2020 Inter Miami CF season was the first season in the history of Inter Miami CF and the 17th season of first-division club soccer in South Florida. It was the first time since 2001 that a South Florida club competed in the first-division. In addition to playing in Major League Soccer, the club was scheduled to participate in the U.S. Open Cup but the competition was cancelled due to the COVID-19 pandemic.

The 2020 FC Cincinnati season was the club's second season in MLS, and the fifth season of a team playing under the FC Cincinnati brand after three years in the lower-division USL Championship. The club finished with a league worst 6–22–6 record in their inaugural MLS season in 2019, setting a league record for the most goals given up with 75. The 2020 season was the final year that FC Cincinnati played home matches at Nippert Stadium, as their new West End Stadium is scheduled to open in March 2021.

Atlanta United played their home games at Mercedes-Benz Stadium. Due to the cancelation of the 2020 U.S. Open Cup, Atlanta competed in the 2021 CONCACAF Champions League as the final berth was given to the defending champions of the U.S. Open Cup.

Outside of MLS, the Union, by winning the 2020 Supporters' Shield, is competing in their first ever CONCACAF Champions League.

New York City FC usually played its home games at Yankee Stadium in the New York City borough of The Bronx. Like the 2020 MLS season, the club also played several of their home games at Red Bull Arena in Harrison, New Jersey due to scheduling conflicts with Yankee Stadium and its backup stadium, Citi Field. New York City FC won the clubs' first MLS Cup title by defeating the Portland Timbers 2–4 in penalty shoot-outs with a 1–1 score before going into extra time, in which neither of the teams scored.
The 2022 season was D.C. United's 27th in existence and their 27th consecutive season in the top division of American soccer, Major League Soccer. In addition to MLS, D.C. United participated in this season's editions of the U.S. Open Cup. United also participated in the Coachella Valley Invitational and the Capital Cup friendly tournaments.
