The enzyme 1-alkyl-2-acetylglycerophosphocholine esterase (EC 3.1.1.47) catalyzes the reaction
The former is also known as platelet-activating factor. There are multiple enzymes with this function:
This enzyme belongs to the family of hydrolases, specifically those acting on carboxylic ester bonds. The systematic name of this enzyme class is 1-alkyl-2-acetyl-sn-glycero-3-phosphocholine acetohydrolase. Other names in common use include 1-alkyl-2-acetyl-sn-glycero-3-phosphocholine acetylhydrolase, and alkylacetyl-GPC:acetylhydrolase. This enzyme participates in ether lipid metabolism.
As of late 2007, 7 structures have been solved for this class of enzymes, with PDB accession codes 1BWP, 1BWQ, 1BWR, 1ES9, 1FXW, 1VYH, and 1WAB.
Platelet-activating factor, also known as PAF, PAF-acether or AGEPC (acetyl-glyceryl-ether-phosphorylcholine), is a potent phospholipid activator and mediator of many leukocyte functions, platelet aggregation and degranulation, inflammation, and anaphylaxis. It is also involved in changes to vascular permeability, the oxidative burst, chemotaxis of leukocytes, as well as augmentation of arachidonic acid metabolism in phagocytes.
In biochemistry, an ether lipid refers to any lipid in which the lipid "tail" group is attached to the glycerol backbone via an ether bond at any position. In contrast, conventional glycerophospholipids and triglycerides are triesters. Structural types include:
Platelet-activating factor acetylhydrolase IB subunit alpha or Lisencephaly protein-1 (LIS-1) is an enzyme that in humans is encoded by the PAFAH1B1 gene. The protein plays an important role in regulating the motor protein dynein.
