Euphorbia resinifera, the resin spurge, is a species of spurge native to Morocco, where it occurs on the slopes of the Atlas Mountains. The dried latex of the plant was used in ancient medicine. It contains resiniferatoxin, an extremely potent capsaicin analog tested as an analgesic since 1997.

Growth

It is a shrub growing to 61 centimetres (24 in) tall, forming multi-stemmed cushion-shaped clumps up to 2 metres (6 ft 7 in) wide. The stems are erect, succulent, superficially like a cactus, four-angled, with short but sharp pairs of 6-millimetre (0.24 in) spines on the angles, spaced about 1-centimetre (0.39 in) apart up the stem. Geographical distribution

Euphorbia resinifera is a species of spurge native to Morocco, where it occurs on the slopes of the Atlas Mountains. [2] It is similar to its relative Euphorbia echinus , which occurs on the Moroccan coast and the Canary Islands. Due to its origin it is also called the African spurge. [3]

Chemical constituents

Euphorbia resinifera contains a milky fluid or latex, which in its dried form is called Euphorbium. It has high concentration of resiniferatoxin, an analog of capsaicin, the primary vanilloid compound found in hot peppers. It can interact with a vanilloid receptor on primary sensory neurons mediating pain (nociception) and neurogenic inflammation. The pain sensing cation channel is TRPV1. [4] Resiniferatoxin has been used as a starting point in the development of a novel class of analgesics. Desensitization to topical resiniferatoxin has been tested in clinical trials to evaluate its potential to relieve neuropathic pain, as in diabetic polyneuropathy and postherpetic neuralgia. [4] Resiniferatoxin injected subcutaneously into a rat hind paw several minutes before a surgical incision reduced postsurgical pain for 10 days in a NIH study published March 2018. [5] It has been tested to treat pain with advanced cancer. [6]

Resiniferatoxin was isolated in 1975. [4] Euphorbium has been used since at least its first written record from the time of Roman Emperor Augustus. [4]

Pliny the Elder states that the name of the drug was given to it in honor of Euphorbus, the physician of Juba II, king of Mauretania. In former times euphorbium was valued in medicine for its drastic, purgative and emetic properties.

Relief from chronic pain remains a recognized unmet medical need. Consequently, the search for new analgesic agents is being intensively studied by the pharmaceutical industry. The TRPV1 receptor is a ligand gated ion channel that has been implicated in mediation of many types of pain and therefore studied most extensively. The first competitive antagonist, capsazepine, was first described in 1990; since then, several TRPV1 antagonists have entered clinical trials as analgesic agents. Should these new chemical entities relieve symptoms of chronic pain, then this class of compounds may offer one of the first novel mechanisms for the treatment of pain in many years.

Zucapsaicin (Civanex) is a medication used to treat osteoarthritis of the knee and other neuropathic pain. It is applied three times daily for a maximum of three months. Zucapsaicin is a member of phenols and a member of methoxybenzenes. It is a modulator of transient receptor potential cation channel subfamily V member 1 (TRPV-1), also known as the vanilloid or capsaicin receptor 1 that reduces pain, and improves articular functions. It is the cis-isomer of capsaicin. Civamide, manufactured by Winston Pharmaceuticals, is produced in formulations for oral, nasal, and topical use.

Euphorbia royleana is a species of flowering plant in the family Euphorbiaceae. It is also known as Sullu spurge, and Royle's spurge. It is a succulent and almost cactus like in appearance although unrelated. It grows right across the Himalaya mountains from Pakistan, India, Bhutan, Myanmar, Nepal to western China, It prefers dry and rocky slopes between 1000 and 1500 meters, but has been found up to 2000 meters. Flowering and fruiting is in spring to early summer (March–July) and seeding is in June–October. It is used as a hedging plant in northern India and has medicinal uses.

Euphorbia abyssinica, commonly known as the desert candle or candelabra spurge, is a species of plant in the family Euphorbiaceae. E. abyssinica is endemic to Ethiopia, Somalia, Sudan and Eritrea. It was first described in 1791, by the German botanist Johann Friedrich Gmelin. In its native habitat, it can grow up to 10 m (33 ft) tall. The woody stem is used for firewood and as timber in roofing, furniture and other items, and the sap is used in traditional medicine. It is also cultivated as an ornamental house plant.
